Human embryonic stem cells (hESCs) are derived from the inner cell mass (rCM) of blastocyst embryos. They are established from surplus embryos that have been obtained by in vitro fertilization (IVF) and donated for research purposes. The rCM-derived cell lines have two unique properties, they can be propagated indefinitely in culture   and have the potential to develop into any cell type in vivo and in vitro. Human ESCs carrying specific mutations   can be used as a powerful tool for studying genetic disorders in human. One preferable approach for obtaining these cell lines is their derivation from affected preimplanration genetic diagnosed (PGD) embryos. These unique   cells are important for modeling human genetic disorders for which there are no suitable research models. They   can be further used to gain new insights into developmentally regulated events that occur during human embryo   development and that are responsible for the manifestation of genetically inherited disorders. In this review, we will introduce a current state of the studies for PGD-derived hESCs carrying mutation for genetic disorders that   have great potential for the investigation of new therapeutic protocols, including gene-therapy-based treatments   and disease-oriented drug screening and discovery.

Human pluripotent stem cells (hPSCs) with the capacity of self-renewal and multilineage differentiation are promising sources for replacement therapies and regeneration medicine. Many researches have been spent to undersrand rhe molecular mechanisms underlying hPSCs mainrenance and pluriporency, and ir is clear rhar borh rranscriprional and epigeneric levels of regularion have crucial roles. Recent research indicares rhar regularorynon-coding RNAs (ncRNAs) are key regulators of hPSCs epigeneric nerwork. For rhis reason, we summarize rhe   findings that focus on regulatory ncRNAs of human embryonic stem cells (hESCs) and human induced pluripotent srem cells (hiPSCs).

Human embryonic stem cells are great resources for cell based therapies and clinical application in regenerative medicine. In this review, we summarize recent developments of culture conditions, including growth medium, extracellular matrix components, physiological environments and maintenance methods in human embryonic stem cells for clinical trials. Based on the researches up to now in human embryonic stem cell culture conditions, the studies should be continued and investigated advances being toward new clinical-grade xeno-free culture systems. Ultimately, it will be more effective production of clinical-grade human embryonic stem cells to useful therapeutic applications in the future.

Regenerative medicine including tissue engineering and stem cells is emerging field for development of cell source for functional recovery of damaged tissue or cells. Tissue engineering of uterus and vagina is essential for the treatment of women with agenesis or dysfunction of female reproductive organs. For this purpose, suitable cells   and biodegradable scaffolds need to be efficiently incorporated into artificial mold of tissues as forms of sheets or layers. In this review, we tried to summarize the recent trends of bioengineering techniques and used materials for the regenerative therapeutic application in female reproductive organs.

Parkinson``s disease (PD) is one of the most common neurodegenerative movement disorder involved in central nervous system. PD is characterized by progressive and selective loss of dopaminergic (DA) neurons in the midbrain substantial nigra. For study of the pathogenic mechanism and development of the therapeutics in PD, various animal models are widely used as an essential tool. PD animal models can be prepared some methods such as neurotoxin administration or genetic modification from normal animals. Suitable animal model must be properly selected according to the purpose of the animal study because each animal model has different specificities and limitations. In this review, we will introduce various animal models of PD and examples of their application.

Age-related macular degeneration (AMD) is characterized by the loss or dysfunction of retinal pigment epithelium (RPE) and one of the major causes of irreversible blindness in the elderly. According to the presence or absence of choroidal neovascu -larization, advanced AMD can be classified into two types (dry AMD and wet AMD). Currenrly, no treatments can reverse dry AMD, regardless of the fact that dietary supplementation with defined vitamins and antioxidants has been shown to alleviate progression. Therefore, RPE replacement represents potential new approaches for dry AMD. Functional RPE cells could be generated from stem cells by spontaneous differentiation, coculture, treatment of the defined factors, or cell reprogramming. In this review, we summarize molecular and structural characteristics of RPE cells and discuss RPE-impairment, RPE-related diseases that will be amenable to cell replacement strategies and several stem cell sources to produce RPE in vitro.

The nervous system is essential for normal physiological function of all systems within the human body, and a complex collection of nerves and specialized cells known as neurons that transmit signals between different parts of the body. However, the nervous system has a limited capacity for self-repair. Recently, stem cell therapy using easily accessible mesenchymal stem cells, has come to as a potential therapeutic regime. Adipose tissue-derived mesenchymal stem cells (ADSCs) have the capacity to differentiate into endodermal and ectodermal cell lineage in vitro. Thus, it is important that not only is it a prerequisite to understand the functional properties of the differentiated cells, but also deciphering the regulatory pathways and mechanisms of the ADSCs themselves, suggesting that it will expedite the development and usage of ADSCs for autologous stem cell therapy in treating neurological diseases. We discuss the neural differentiation potential of human ADSCs and potential implication of these cells in treating neurodegenerative diseases.

Platelets are known as a natural source of various growth factors which contribute to cell activation and tissue regeneration. These released growth factors induce chemotaxis, cell proliferation, differentiation, and angiogenesis, and play a role in wound healing and regenerative process. Platelet rich plasma (PRP) is a concentrate of platelets. Except of platelets, substances needed for regeneration like fibronecrin, vitronectin are included in PRP. Thus, clinical application of PRP could be a part of promising therapy methods due to combination effects of various factors in PRP. A possible role of platelets in cartilage regeneration has been reported. Researchers found convincing evidences for activation of chondrocyte proliferation and increase of proteoglycan production. Recently, microRNAs were discovered that function as an inducer for differentiation of mesenchymal stem cells (MSCs) into chondrocytes. In this study, anti-inflammatory and regeneration effects of PRP in cartilage disease will be discussed.

Immune checkpoint receptors are crucial molecules for adjusting immune responses. Checkpoint signaling dampens T cell activation to avoid autoimmunity and the destructive effects of an excessive inflammatory response. It is well established that tumors use several mechanisms to avoid elimination by the immune system. Checkpoint blockade therapy utilizes monoclonal antibodies to release the brakes from suppressed T cells, allowing them to be activated and recover their antitumor activity. This therapeutic approach has revolutionized cancer immunotherapy, and extraordinary increases in overall survival were noted, first with anti-CTLA-4 (cytotoxic T lymphocyte associated protein-4) and subsequently with anti-PD-l (programmed cell death receptor-I) in melanoma and other malignancies. The complex biology of immune checkpoint pathways still contains many mysteries, and the full activity spectrum of checkpoint blocking drugs is currently the subject of intense study. In this review, we discuss the discovery of immune checkpoints, the clinical application of their inhibitors and the future directions of this highly interesting class of molecules.

During the last 3 decades, oncogenic Ras has been recognized as one of the potent causes of human cancer. But until now, there is no safe and effective Ras inhibitor, because alternative pathway is turned on when Ras inhibitor is treated. So there were many attempts to inhibit the post-translational modification termed farnesylation, which is essential for Ras anchorage into the membrane. Previous attempts to target farnesylation have focused on the development of farnesyltransferase inhibitors, but the performance of such agents in cancer clinical trials has not been as good as hoped. Here, we review novel therapeutic approaches targeting prenylation and prospect the future of using prenylation inhibitors as cancer therapy
