In this study, a carnitine salicylate ion pair (called IP-BHA) was prepared through hydrogen bonding and dipole-dipole interaction, which are known to possess exfoliating and anti-inflammation properties. In an in vitro study, the efficacy of carnitine salicylate on keratin exfoliation at weakly acidic pH and cytotoxicity and alleviation of the pro-inflammatory factor nitric oxide was superior to that of salicylic acid. As a result of analyzing the degree of keratin exfoliation using a colorimeter in the clinical evaluation of keratin peeling performed on the forearms of 12 participants aged 20 ∼ 30 years, the IP-BHA (0.5% salicylic acid) applied experimental group improved by 76.37 ± 9.16% after 14 days compared to the vehicle applied group. Erythema mitigation effect was evaluated by inducing erythema using applying 1% sodium dodecyl sulfate solution to the forearms of 9 participants aged 20 ∼ 30 years for 24 hours. As a result, the IP-BHA applied g roup improved by 74.03 ± 14.82% a f ter 12 days compared to the vehicle applied g roup. Through sensory evaluation, it was verified that the skin texture was improved and skin soothing was felt by IP-BHA. This study indicates that IP-BHA can be used as a cosmetic material with excellent effects on exfoliation and erythema relief.

The stratum corneum, which exists in the outermost layer of the skin, has a skin barrier function that protects the skin from harmful substances. In addition to the degree of differentiation of keratinocytes and the composition of intercellular lipids, corneocyte lipid envelope which is covalently bound to cornified envelope is also important to be associated with skin barrier functions. Some of corneocyte isolated from stratum corneum would be stained by bradford reagent. The bradford staining ratio of corneocyte showed a positive correlation with age and trans epidermal water loss. It was also confirmed that some Staphylococcus aureus (S. aureus) was more attached to the corneocyte stained with bradford reagent. Natural extracts that reduce the staining ratio of corneocyte suppresses trans epidermal water loss and inhibits the adhesion of S. aureus on stratum corneum. In conclusion, bradford staining of corneocytes could be used as a method to evaluate skin barrier function for both moisturizing and antibacterial. And this evaluation of staining corneocyte could be used as a method of selecting materials that improve the skin barrier function.
