In this study, the fungal strain KNUF-21-F39 was isolated from a declined apple tree (Malus domestica) in the Chungcheongbuk province in Korea. The strain KNUF-21-F39 presented a slow growth rate and a variety of macroconidia shapes and sizes ranging from ovoid to fusoid and 1- to 5-septate, primarily showing 3- and 4-septate, with “S”-shaped macroconidia rarely observed. The strain was identified based on morphological characteristics along with phylogenetic analysis performed using the internal transcribed spacer region (ITS) and partial sequences of translation elongation factor 1-α (tef1), RNA polymerase largest subunit (rpb1), and calmodulin (cal) genes. The fungal strain KNUF-21-F39 was identified as Fusarium diversisporum, which has not been previously reported in Korea. The ice nucleation activity (INA) of the strain was also evaluated, identifying the strain as positive for INA. This is the first report characterizing F. diversisporum as an IN-active fungal species.

Lentinula edodes is an important commercial mushroom and there have been several reports of viral infections in L. edodes. Two mycoviruses (LeV-HKB and LeNSRV1) were detected in Sanbaekhyang (NIFoS 2778) and Taehyanggo (NIFoS 4317), the sawdustcultivated commercial strains. The vertical transmission rates of the viruses were investigated by detecting the viruses in 80 monokaryotic strains derived from basidiospores isolated from the fruiting bodies of each strain. Most of the monokaryotic strains were infected with the virus and the two viruses showed different levels of meiotic stability, with LeV-HKB showing higher meiotic stability than LeNSRV1. Therefore, it seems that the vertical transmission mechanism of mycoviruses is different depending on the virus species. We also examined the mycelial growth rate of the monokaryotic strains and compared the growth rate according to virus infection status. Although there was no statistically significant correlation between viral infection and mycelial growth rate, we found that the average growth rate was reduced by additional virus infection. We expect our data to contribute to a greater understanding of the mechanism of the vertical transmission of mycoviruses, and promoting breeding using virus-free monokaryotic strains.

Truffles are ectomycorrhizal fungi that belong to the genus Tuber. They exhibit symbiotic relationships, particularly with oak (Quercus spp.) and hazel (Corylus spp.) trees. We performed an inoculation using a spore suspension to synthesize mycorrhizae between European truffles, Tuber borchii and Tuber melanosporum, and an indigenous oak species, Quercus acutissima. This resulted in the formation of mycorrhizae within 2 months after inoculation. Despite having the same host plant, differences in features were observed between Tuber species, including color and mantle type. These results indicate that Q. acutissima is a suitable host plant for truffle cultivation in Korea and provide a better understanding of the mycorrhization of T. borchii and T. melanosporum.

Fusarium head blight is an important disease of small grains. It is mainly caused by members of the Fusarium graminearum species complex (FGSC). Barley and wheat growers spray fungicides, especially demethylation-inhibitor fungicides, to suppress the disease. The objective of this study was to examine the changes in the sensitivity of the FGSC population to the triazole fungicide, propiconazole. A total of 124 and 350 isolates of FGSC were obtained from barley and wheat in Jeolla Province during 2010-2016 and 2020-2021, respectively. The species identity and trichothecene chemotypes of the FGSC isolates were determined based on polymerase chain reaction assays targeting translation elongation factor 1-alpha and TRI12 genes, respectively. Sensitivity to propiconazole was determined based on the effective concentration that reduced 50% of the mycelial growth (EC50) using the agar dilution method. Of all isolates, F. asiaticum with the nivalenol chemotype was the most common (83.9% in 2010-2016 and 96.0% in 2020-2021), followed by F. asiaticum with the 3-acetyl deoxynivalenol chemotype (12.1% in 2010-2016 and 2.9% in 2020-2021). The EC50 values of the isolates collected in 2010-2016 and 2020-2021 ranged from 0.0180 to 11.0166 μg/mL and 1.3104 to 17.9587 μg/mL, respectively. The mean EC50 value of the isolates increased from 3.8648 μg/mL in 2010-2016 to 5.9635 μg/mL in 2020-2021. The baseline resistance to propiconazole was determined to be 7 μg/mL, based on the EC50 value of isolates collected in 2010-2016, and the ratio of resistant isolates increased from 9.7% in 2010-2016 to 28.6% in 2020-2021.

In this study, the physical properties of the medium and changes in the wood chemical composition of the sawdust were investigated during the cultivation of oak mushroom sawdust bags, and the following results were obtained. After inoculation, the weight of the medium decreased during the incubation period. It is determined that this is not due to evaporation of moisture containing the medium or decomposition of sawdust, but to decomposition of rice bran, a low molecular substance added to the medium. It was confirmed that the moisture content of the medium was steadily increased during incubation, and it was estimated that the organic substrates such as rice brane in the medium was decomposed by mycelium, and water, one of the decomposition products of organic substrates, caused an increase in the moisture content of the medium. Along with the increase in the harvest of oak mushrooms, the proportion of organic substances such as holocellulose and lignin, the main components of the wood cell wall of sawdust, steadily decreased. In particular, the degradation characteristics of the wood cell wall component of shiitake, which is a white rot fungi, were confirmed by higher lignin reduction rate than that of holocellulose. On the other hand, ash, which is an inorganic material, increased with an increase in the number of mushroom harvests. The increase in the amount of ash in the medium may have been due to the decrease in the organic matter content such as holocellulose and lignin.

Lentinula edodes is a type of mushroom widely consumed in East Asia, and many of the components contained in them are used as raw materials for pharmaceuticals or as health supplements. Ergothioneine, a sulfur-containing amino acid in L. edodes, is attracting attention due to its antioxidant and anti-inflammatory effects. L. edodes generally contain an ergothioneine content of about 200 to 800 mg per kg; there are some differences in this value depending on the cultivation conditions. In this study, 24 domestic sawdust-cultivated L. edodes cultivars were cultured under the same conditions, and the morphological characteristics and ergothioneine content of the fruiting bodies were investigated. The yield and morphological characteristics of the fruiting bodies were different in each cultivar. Taehyanggo and Sanjo 713ho had the largest and thickest pileus. The pileus of L. edodes had a higher ergothioneine content than the stipe. Sanjo 715ho had the highest ergothioneine content in the pileus and the stipe at 1,225 mg/kg and 753 mg/kg, respectively. An ergothioneine content of more than 1,000 mg/kg in the pileus was observed in Bambithyang, Sansanhyang, Sulbaekhyang, Taehyanggo, Sanjo 705ho, Sanjo 709ho, Sanjo 715ho, and Sanjo 716ho. These cultivars can be used as parental strains to develop new cultivars with enhanced ergothioneine functionalities.

Endophytic fungal strains were isolated from the leaves of two conifer species (Juniperus rigida and Pinus densiflora) in Korea and identified on the basis of their morphological and molecular characteristics. Internal transcribed spacer and large subunit regions of rDNA were used for the phylogenetic analysis, and the translation elongation factor 1-alpha (TEF) and RNA polymerase II second largest subunit (RPB2) genes were analyzed depending on the species. Two fungal species that were previously unrecorded in Korea were identified: Acrodontium burrowsianum and Pestalotiopsis humicola. Their morphological and phylogenetic characteristics are described herein.

In July 2020, wilting symptoms were observed among adzuki bean plants (Vigna angularis var. angularis L.) in the fields in Yeosu, Korea. Infected plants showed yellowing of leaves, browning inside the stems, splitting of stem bark, and wilting. When these plants were uprooted, their roots were found to be brown. The fungal pathogens NC20-737, NC20-738, and NC20-739 were isolated from symptomatic stem and root tissues. These pathogens were identified as a Fusarium fujikuroi species complex based on their morphological characteristics. Molecular identification was performed using the DNA sequence of translation elongation factor 1 alpha and the RNA polymerase Ⅱ second largest subunit regions. The nucleotide sequences of all three isolates were similar to the F. fujikuroi reference isolates NRRL 13566 and NRRL 5538 of the National Centre for Biotechnology Information GenBank. A pathogenicity test was conducted by the soil inoculation method with cornmeal sand inoculum. Approximately 3 weeks after inoculation, symptoms were observed only in the inoculated adzuki bean seedlings. To the best of our knowledge, this is the first report of Fusarium root rot caused by F. fujikuroi in adzuki beans, both in Korea and worldwide.

This study was conducted to identify yeast species isolated from domestic pear blossom through gene sequencing and analysis of morphological characteristics, and to confirm specific yeast species inhibitory effects toward fire blight in immature apples, pears, and crab apple blossoms. Yeast morphological characteristics were consistent with the known characteristics of Aureobasidium pullulans. Nucleotide sequencing of the D1/D2 region of large-subunit (LSU) 26S ribosomal DNA and the internal transcribed spacer (ITS) region confirmed its identity as A. pullulans (MHAU2101). Inoculation of immature fruits with A. pullulans MHAU2101 before exposure to Erwinia amylovora prevented fire blight symptoms in apples and pears. A. pullulans MHAU2101 treated crab apple blossoms had a significantly lower flower infection rate than untreated blossoms, revealing 64% of the potency of streptomycin. The A. pullulans MHAU2101 treated group also displayed lower E. amylovora density in both pistil and hypanthium compared to the untreated group, especially in the hypanthium. This study confirms that A. pullulans MHAU2101 isolated from domestic pear blossom can effectively suppress the onset of fire blight.

Following the taxonomic reassignment of Schizopora and some Hyphodontia species into Xylodon, we confirmed nine Korean species of Xylodon in a previous study. In the present study, we propose Korean names for these Xylodon species according to the Principles of the Mycological Terminology Review Committee of The Korean Society of Mycology.
