1Seven Newly Recorded Macrofungi of Inocybaceae (Agaricales, Basidiomycota) in Korea

The Inocybaceae is a family of ectomycorrhizal fungi belonging to the Agaricales order. Most species in this family produce secondary metabolites, such as muscarine, psilocybin, and other compounds. In the present study, we conducted taxonomic studies on the Inocybaceae based on morphological and molecular phylogenetic analyses for the first time in Korea. As a result, seven species of Inocybaceae were newly recorded: Inocybe caroticolor, I. fuscidula, I. grammatoides, I. latibulosa, I. stellata, Inosperma shawarense, and Mallocybe malenconii. Among them, the genera Inosperma and Mallocybe were described for the first time in Korea. Relevant morphological characteristics, phylogenetic trees, and photographs are provided.

Collema Weber ex F.H. Wigg is scarcely scattered throughout South Korea. However, a detailed taxonomic or revisionary study has not been carried out for almost two decades. The present study was based on specimens deposited in the lichen herbarium at Korean Lichen Research Institute, which were identified using recent literature. In this revisionary study, a total of sixteen species of Collema, including five new records, C. furfuraceum var. luzonense, C. pulchellum var. subnigrescens, C. rugosum, C. subconveniens, C. tenax, and one variety C. leptaleum var. leptaleum were documented. Further, detailed descriptions of each species and their morphological, anatomical and chemical characteristics and a key to all known Collema species of South Korea were reported.

3Phylogeny and Morphology of Sarcopodium terrigenum sp. nov., a Novel Fungal Species Isolated from Soil in Korea

A fungal strain was isolated from a soil sample collected in Korea and designated as YW23-8. Based on a sequence analysis of the internal transcribed spacer (ITS) regions, the isolate was assigned to the genus Sarcopodium. Moreover, a phylogenetic analysis based on the concatenated nucleotide sequences of the ITS regions and the large subunit of the nuclear ribosomal RNA (LSU) gene showed that the strain YW23-8 occupies a distinct phylogenetic position within Sarcopodium . The isolate had significant differences from its closest neighbors, S. circinosetiferum, S. circinatum, S. macalpinei, and S. vanillae. Morphological features such as different conidial structures, the absence of septation in conidia, and the presence of milky white watery droplets along with the results of the phylogenetic analysis clearly distinguish YW23-8 from the closest Sarcopodium species. We therefore conclude that strain YW23-8 represents a novel species of the genus Sarcopodium for which we propose the name Sarcopodium terrigenum.

Various freshwater ecosystems, such as streams, lakes and rivers, provide a dynamic habitat for fungi. In this study, we isolated several fungal strains from freshwater sediment and plant litter. The strains were identified using molecular phylogenetic analyses of rDNA and beta tubulin (TUB) sequences. Morphological characteristics of the fungi were also investigated using microscopy and culture characteristics of the fungi grown on several media. We identified seven species previously unrecorded in Korea, Nothophoma spiraeae, Westerdykella aquatica, W. aurantiaca, W. dispersa, Chrysosporium sanyaense, C.pseudomerdarium and Taeniolella phialosperma.

The present study was performed to improve the technique used for fermenting the mushroom growth medium. Taxonomic analysis of 16S rDNA sequence from the predominant Bacillus strain CY-24 isolated during the fermentation phase of the rice straw medium identified it as Bacillus licheniformis. In addition, the growth environment of B. licheniformis was also examined in this study, which revealed the optimal growth temperature and pH to be 30 ℃ and 6.0, respectively. This study also revealed that carboxymethyl cellulase (CMCase) and polygalacturonase (PGase) enzymes isolated from B. licheniformis achieved their maximal activities at 50 ℃ and 60 ℃ respectively. Furthermore, the study confirmed that the two enzymes, i.e., CMCase and PGase in B. licheniformis are stable at temperatures above 60 ℃. The present study thus demonstrates that B. licheniformis CY-24 possesses excellent enzymatic properties. It also reveals that the action of enzymes during the production of growth mediums used for the cultivation of mushrooms is closely associated with the promotion of fermentation and softening of the rice straw. Overall, this study provides elementary information regarding the role of B. licheniformis enzymes during growth medium fermentation for Agaricus bisporus cultivation.

Indigenous yeasts in Korea have been actively studied since 1910 in light of various fields, including environment, food fermentation, brewing, and medicine. However, a complete list of indigenous yeast species has not been addressed or reported. In this study, a comprehensive analysis of the literature including yeast isolation from Korean materials and the strains preserved at culture collections was performed. The results of this study indicate the following: ⅰ) there are 128 genera and 681 species (including 26 variants) of yeast reported in Korea, ⅱ) the reported species were trimmed into 142 genera and 500 species (including 9 variants), ⅲ) among which only 166 species were registered in the National Species List of Korea (2020). Therefore, follow-up studies are required to register more indigenous yeast species in the National Species List of Korea.

In this study, screening of potent non-pathogenic wild yeast with high anti-dementia butyrylcholinesterase (BChE) inhibitory activity and production condition of a BChE inhibitor were described. Among 36 non-pathogenic wild yeasts, Saccharomyces cerevisiae WJSL 0113 showed the highest BChE inhibitory activity of 85.2%. The specific BChE inhibitor was maximally produced when S. cerevisiae WJSL 0113 was cultured at 30℃ for 48 h in a yeast extract-peptone-dextrose medium.

8Damping-off of Deltoid Synurus Caused by Rhizoctonia solani AG-1 (IB)

Damping-off symptoms were frequently observed in young deltoid synurus (Synurus deltoides) plants grown in a farmer's vinyl greenhouse located in Hwacheon, Gangwon Province, Korea, during a disease survey in July 2019. The incidence of diseased plants in the vinyl greenhouse investigated was 5-30%. Five isolates of Rhizoctonia sp. were obtained from the diseased plants and identified as Rhizoctonia solani AG-1 (IB), based on morphological and cultural characteristics and anastomosis test results. Three isolates of R. solani AG-1 (IB) were tested for pathogenicity on deltoid synurus using artificial inoculation. All the tested isolates induced damping-off symptoms in the inoculated plants. The symptoms were similar to those observed in plants from the farmer's vinyl greenhouse. To our best knowledge, this is the first report of R. solani AG-1 (IB) causing damping-off in deltoid synurus.

9Puccinia klugkistiana, a Rust Fungus Occurring on Cleistogenes hackelii in Korea

Rust samples at both uredinial and telial stages were collected from Cleistogenes hackelii (Poaceae) in 2017 and 2019 in Korea. Based on the morphological examination and molecular phylogenetic analyses of the internal transcribed spacer (ITS) and large subunit (LSU) rDNA, it was identified as Puccinia klugkistiana. This rust at aecial stage has previously been recorded on Ligustrum japonicum (Oleaceae) in Korea. Puccinia diplachnicola has been previously recorded on C. hackelii in Korea; however, it is believed to be unrelated to the rust collected in the present study. Conclusively, this is the first study to report P. klugkistiana occurrence on C. hackelii in Korea.
