Induction of Plantlets by Bud Culture in Quercus acutissima Institute of Forestry Genetics :

To establish a vegetative propagation method Quercus acutissima axillary buds with stem segment collected from 2-monthold seedlings were cultured on MS, MMS, 1/2MS, WPM and GD agar media containing BAP, Kinetin, NAA and IBA. The results obtained were as follows; 1. Axillary buds were successfully induced (71-96%) within 30 days on MS medium containing only BAP 0.5 to 5ppm and on MMS, 1/2 MS media containing NAA 0.05ppm in combination with BAP 0.5 to 5ppm. Best result was obtained on MMS medium containing 1.0ppm BAP and 0.05ppm N.AA (96%) When shoots developed from initial explants were excised into single shoots(1-1.5㎝) and cultured on various media(MS, MMS, WPM) containing BAP(0.1-3.0ppm) and NAA(0.01-0.5ppm) for 30 days, best result was obtained on MS medium supplemented with 0.8ppm BAP and 0.01 NAA (7.6 shoots). Of the media tested, MMS medium containing 0.05ppm BAP and 0.5ppm NAA was the best for shoot growth (5.9㎝). 3. Root initiation was successfully developed on GD medium containing 0.2ppm IBA(76.7%) within 4 weeks.
