The current study was undertaken to determine whether recombinant porcine growth hormone(ppGH) and pituitary porcine growth hormone(ppGH) inhibited lipogenesis in the porcine adipose tissue. Backfat tissues were obtained by biopsy from 6 barrows. The adipose tissue explants were cultured in the M199 for 48 hrs with various concentrations of pGH (0, 0.1, 1.25, 1, 5, 10, and 100 ng/㎖) before they were incubated in the KRB buffer for 2 hrs for lipogenesis measurement. Lipogenesis was determined as glucose conversion to total lipid. Fifty ng/㎖ of hydrocortisone and 10 ng/㎖ of insulin were included in the culture media and incubation buffer to induce maximum pGH effect. Recombinant pGH and ppGH similarly inhibited lipogenesis in a dose-dependent manner, (EC]_(50) being 0.57 ng/㎖ for both LrGHs. Inhibition of lipogenesis by both pGHs was considerable, with a 93% reduction at the 5 ng/㎖concentration. These results partly explain the mechanism by which pGH administration reduces carcass fat in swine.
