Five different raisin production media have extensively tested to choose the best: for raisin synthesis by Streptococcus lactis IF012007. S. lactis IF012007 produced 1830 IU/㎖ of total raisin activity and 36.24% of the total nisin activity was cell-bound in the Nat S04-supplemented LTB medium. In the pH-controlled culture, S. lactis IF012007 produced 2350 IU/㎖ of total nisin activity and 94.46% of the total raisin activity was cellbound. In automatically pH-controlled culture, S. lactis IF012007 produced 260() IU/㎖ of total raisin activity and 95.3% of total raisin activity was cell-bound. The crude raisin extracted by hot extraction method with 0.05N HCl was purified by column chromatography employing Amberite resin CG-400 and CM-Cellulose. The purified raisin was found to be homogeneous on polyacrylamide gel electrophoresis.
