To measure the specific activity of liver glycogen some preliminary experiments were carried out. The liver samples from a starved chicken and standard glycogen were digested with hot potassium hydroxide. The glycogen was precipitated with sodium chloride and ethanol. The hydrochloric acid was treated for two and half hours to hydrolyze the glycogen. In order to remove nitrogenous material various precipitants were compared. Ba(OH)₂-ZnSO₂ revealed the best result for the radioassay. However, chemical recovery was 68% in average through this procedure, this value was thought to be derived from free(except fixed) glycogen. Cellosolve(2-ethooxyethanol) showed more accurate counting results than ethanol as blendor for aqueous glucose solution brought from above procedure, using diotal scintillator. The absolute concentrations of glycogen were 0.9 to 1.1%, in the liver samples taken from starved chicken.
