Studies on Increasing Feed Value of Rice Hull (Ⅱ . Some Measurements of Cellulolytic Activities of Fermented Feed - Rice Hull )

This experiment was carried out to measure the cellulolytic activities of fermented feed-Rice Hull-which was cultured with Rice Hull Meal and Wheat Bran at the rate of 4 : 1 and 3 : 2 and also the optimum conditions in various pH ranges were measured and the results were compared to select the best mold having high cellulolytic activity among the employed 6 different molds. In this study the following activities were observed and measured; (1) Cellulolytic Activities for the insoluble fiber; a. Using filter paper as substrate, the disintegration of filter paper sheets(1×1㎝) were observed. b. Using filter paper strip as substrate, the produced amount of glucose was measured by Somogy`s procedures. (2) Cellulolytic Activity for the soluble fiber; A soluble cellulose derivates, Sodium salt of Carboxy Methyl Cellulose was used as substrate of cellulolytic enzymes. And measured the reductivity by Somogy`s procedures. (3) Cellulolytic Activities for the Salcine; Paranitro Phenyl B-D-Glucoside, Salcine was used as substrate of cellulolytic enzymes and the Somogy`s procedures were employed to measure the produced amounts of glucose. From the above measurements of cellulolytic activities of enzymes following results were obtained. 1. The cellulolytic activities of enzymes having strong activities for the insoluble fiber, the highest activities for the complete breakdown of substrate(disintegration of filter paper tissue), were 02∼1, and U-No 2 molds in pH 4 and 5 at the rate of both two mixtures. But the mold, Japonicus, shown the highest activities in pH 4 only at the rate of 3 : 2 mixture. 2. The highest saccharinative activities for the insoluble fiber among the employed molds shown 1.4㎎/5㎖ of Glucose in pH 5 at the rate of 4 : 1 mixture of 02∼1 mold. Generally, the activities shown at the rate of 4 : 1 mixtures were superior to that of 3 : 2 mixtures even in different pH ranges. 3. The highest saccharinative activities for the soluble fiber among the employed molds shown in pH 5 and 6 even at the different rate of mixture. And about 20% of substrates in weight were saccharinated in figuring that activities shown 0.98㎎/㎖ of glucose in the case of U-No2 mold at the rate of 4 : 1 mixture. 4. The saccharinative activities of U-No2, 02-1 and Japonicus were measured as 11%, 9.4% and 9.4% respectively in figuring the produced amounts of glucose. Assuming from the above results the activity of enzyme of the best mold raight be increase in proportion to the concentration of enzyme and the concentration of substrates and its holding hours.
