Biosynthesis of 1,12-dodecanediol and indigoid bio-pigments using Cytochrome P450 monooxygenase biocatalysts

In this presentation, two examples of CYP-dependent whole cell biotransformation of 1-dodecanol and indole-derivatives were demonstrated. First, CYP dependent biosynthesis of 1,12-dodecanediol was investigated through 1-dodecanol oxidation. E. coli cells over-expressing CYP153A33 coupled with putida redox and FadL could produce 173 mg/L of 1,12-dodecanediol under the optimal reaction conditions. Secondly, CYP102G4-dependent oxidation of indole-derivatives involved in novel indigo pigment synthesis was demonstrated in the presentation as another CYP application. Expression of CYP102G4 with the supplement of indole-derivatives resulted in the generation of various colored indigoids with antibacterial activity. These results demonstrated the versatile application of CYP biocatalysts for biosynthesis of polymer monomers and functional bio-pigments, which can be further applied to functional bio-polymer industries.
