Tuning of coenzyme B12 riboswitch sensor by using toehold switch based genetic modulator

Genetic circuits have been widely utilized to control the function of microbial cells, and riboswitches can be adopted as an input part for genetic circuits. The tuning of dose-response parameters is important for diverse applications in synthetic biology. However, the typical tuning approaches for the riboswitches were constrained by their difficulties and narrow range of modulation. Therefore, a new modular part is required that can tune the dose-response parameters with ease. Here, we developed a toehold switch-based genetic modulator. The previously reported hybrid input part and toehold switch were combined, and the resulting genetic circuit showed dramatic improvements in the dynamic range and operational range. Furthermore, the modularity of the toehold switch was also demonstrated using an inducible promoter system. This strategy is applicable for tuning other biosensor systems with ease and could be widely implemented in metabolic engineering and synthetic biology.
