Tobacco Leaf Ethanol Extract Lower Blood Glucose and Decrease Body Weight along with Food Intake in Type 2 Diabetes Mellitus (T2DM) Rat Model

Objective: In T2DM, it is very important to control blood sugar levels, body weight and food intake for preventing the complications of DM. Recent studies show that nicotine can reduce oxidative stress, excessive inflammation and increase insulin secretion. Besides, nicotine also reduces weight through dependent and independent food intake. This research aimed to explore the effect of tobacco leaf extract on T2DM animal model. Methods: This study examined the effect of Ethanol extract of Nicotiana tabacum leaves on the glucose levels, bodyweight, and food intake of diabetic Wistar rats. Male 8-10 weeks old rats were used in this research. Diabetes was induced in rats by high fat diet for 16 days and administration of 22.5 mg streptozotocin/kg body weight intraperitoneally. Fasting Blood Glucose were measured and criteria used for diabetes was fasting blood glucose above 180 mg/dL. Four groups of diabetic rats and one group of normoglycaemic rats (n = 5) were used for the study. Sham and STZ-HFD groups served as non-diabetic and diabetic controls respectively. P1, P2, and P3 groups received tobacco leaf extract of 90, 180, 270 mg/kg body weight respectively for 27 days. Weekly blood glucose level and body weight changes measurement were done along with daily food intake checking. Results: Administration of the extract at all doses showed significant reduction in fasting blood glucose level compared to STZ+HFD group (Mann Whitney test, p = 0.009) on day 27. Food intake is significantly decreased in all days of treatment (Mann Whitney test, p < 0.05) in P3 compared to STZ-HFD group. Body weight of rats were gradually lower following the increment of the dose. Conclusion: The Tobacco Leaf Ethanol Extract might be useful in ameliorating diabetes' sign and symptoms. But further research is needed to determine the effect of extracts on weight loss in DM; whether it's good or bad.
