Background: Psoriasis is a chronic inflammatory skin disease. Differential expressed gene analysis for psoriasis patient can provide insights on pathophysiology Objectives: To elucidate the various and specific genes related to psoriasis pathogenesis by performing differential expressed gene analysis. Methods: Total 7 patients who were histologically diagnosed with psoriasis were enrolled. All 7 patients are severe psoriasis patients(PASI>10). Psoriasis lesional and non-lesional skin biopsy tissues were obtained. Total RNA sequencing was done by using TruSeq Stranded Total RNA Sample Prep kit (Theragen, Seoul, Korea). We did differential gene expression analysis. Finally, we investigated the pathways that are associated psoriasis using gene set enrichment analysis. Results: DEG analysis revealed significant fold-change in KRT 16, KRT6A, SERPINB4, DEFB4A genes of Korean psoriasis patients. Geneset enrichment analysis showed several pathway. Conclusion: These DEG analysis and geneset enrichment analysis can provide understanding about pathophysiology and specific genes
