Synergistic hair growth promoting effects of minoxidil and electrical stimulation in human dermal papilla cells

Background: There have been clinical studies demonstrating the positive effect of electrical stimulation (ES) on hair growth. No previous study showed a synergistic effect of minoxidil (MXD) and ES on hair growth. Objectives: We aimed to investigate the effect of MXD combined with ES on human dermal papilla cells (hDPCs) in vitro. Methods: Cultured hDPCs contained 0.1レM MXD solution were electrically stimulated with two differentvoltage powers (3.5V, 8V) at 1MHz alternating-current for 10 min. After stimulating hDPCs, cell proliferationwas measured by MTT assay. The モ-catenin expression was measured by western blot and immunofluorescence assay. In addition, MAPK pathway such as ERK and AKT levels were measured by western blot assay. Wnt/モ -catenin pathway related genes were measured by real time-PCR. Results: Cell proliferation increased by 61% in 3.5V-1Mhz-MXD group compared with untreated control. Wnt/モ-catenin related gene (Wnt5a, モ-catenin, Lef1 and Axin2) expression increased in specific frequency condition and モ-catenin was strongly expressed in nuclear localization under ES with MXD groups compared with ES only groups. Likewise, ERK and AKT protein levels increased in ES with MXD groups compared with ES only groups. Conclusion: MXD combined with ES has a synergistic effect on hDPCs proliferation. This might be through activation of Wnt/モ-catenin pathway and MAPK pathway.
