Bone Marrow Stromal Cell Adhesion of Type I Collagen-Coated Scaffold Material: Scanning Electron Microscopic and Histologic Evaluation

The scaffold surface can be altered by surface coating materials. We evaluated the effect of a collagen type 1 coating on the scaffold material(Bio-Oss block bone(R), Geistlich, Swiss). Cell adhesion on the scaffold of mongrel dog bone marrow stromal cells(dBMSC) obtained from iliac bone was evaluated via scanning electron microscopy(SEM) and histologic evaluation. dBMSC were cultured for 1-,2- and 3-week for SEM. And two mongrel dogs(17 and 19 kg of weight, about 2 years old) were used in this study. Right and left mandibular premolars were extracted. 2 saddle type defects(1×1×1 cm) were made at premolar areas 2-month after surgery, and then sacrificed at 6-week after surgery for histologic evaluation. The resin blocks were sectioned in 7 ?m thin slice for villanueva bone stain. In SEM findings, the surface of collagen-coated scaffold materials shows smoother than that of collagen non-coated materials, and the dendritic shape characteristic of cell adhesion with the lapse of culture time, especially 2-week culture. And in histologic findings, the collagen-coated scaffold materials shows more osteocytelike cells on the matrix than collagen non-coated materials. This study suggested that an scaffold materials coated with collagen type 1 stimulated the adhesion of dBMSC. In the future, the research is needed to examine implant placement at the vertical alveolar ridge augmented area in vivo.
