Inducement of Chondrogenic Differentiation in Injectable Hydrogels Embedded in Mouse Embryonic Stem Cells and Growth Factor for

Injectable hydrogel with growth factor was fabricated and investigated as a delivery vehicle of mouse embryonic stem cells(mES) for neocartilage formation. To determine the optimal growth factor in injectable hydrogel, we evaluated the GAG/DNA assay, RT-PCR analysis, and histology. GAG/DNA assay showed that TGF-β 3 enhanced the secretion of GAG content and proliferated well comparing to without TGF-β 3. RT-PCR results for the resultant cartilage tissue revealed that p(NiPAAm-co-AAc) hydrogels with TGF-β 3 was optimal for cartilage tissue formation in terms of collagen Type II and COMP gene expression and showed the results of histology and immunochemical histology.
