양수에서 AChE검사시 양수에 오염원으로 작용할 수 있는 FBS, BCS, 모체와 태아의 혈장 및 적혈구 세포에 의한 위양성 가능성을 전기영동방법으로 확인하여 다음과 같은 결과를 얻었다. FBS는 여러 오염물질중에서 가장 강력한 양성반응을 보였으며 0.01% 이하의 농도에서도 양성을 보일만큼 AChE가 다량으로 존재하였으며, 그 성상도 신경관결손증에 특이한 AChE와 유사하였다. 반면에 BCS와 태아 및 모체의 혈장의 경우는 5%이상의 농도에서도 모두 음성반응을 보였다. 한편 모체의 적혈구세포는 수용성 분획과 세정제(Triton X-100) 용해성 분획 모두에서 그리고 태아의 적혈구세포는 세정제 용해성 분획에서 신경관결손증에 비특이적인 대단히 이질적인 AChE가 다량으로 검출되었다. 따라서 이들 분획중 세정제 용해성 분획은 일반적인 AChE검사에서 세정제를 사용하지 않는다는 점을 고려할 때 위양성으로 작용할 가능성은 희박하나, 모체 적혈구세포의 수용성 분획은 주의를 기울이지 않으면 경우에 따라 위양성으로 오인될 가능성이 매우 높은 것으로 나타났다.

The effects of contamination on the AChE gel electrophoresis assay used in the diagnosis of NTDs were investigated with aniotic fluids artificially contaminated with FBS, BCS, maternal and fetal blood, respectively. The amniotic fluids contaminated with FBS showed strong false positive results below 0.5% concentrations and its profile was nearly identical to that of NTDs-specific AChE. Whereas BCS and fetal and maternal plasmas gave negative results at all concentrations ranging from 5% to 0.5% contaminations. The water-soluble fraction of maternal erythrocytes and the detergent(TritonX-100)-soluble fraction of both of maternal and fetal erythrocytes were elucidated to contain a significant amount of AChE that illustrated a very heterogeneous pattern of fast moving bands located slightly different positions compared to NTDs-spedcific AChE. It was, thus, suggested that, though the detergent-soluble fraction had little posibility of acting as false positive on considering that normla AChE assay procedures exclude any dertergent including Triton X-100, the water-soluble fraction might yield false positive results occasionally unless careful careful exmination was not taken.
