사람태반 양막 및 융모막 조직내의 Diamine Oxidase 활성도에 관한 연구
Study of Diamine Oxidase Activity in Human Amniotic and Chorionic Tissues

사람의 양막과 융모막 조직에서 핵분획, 미토콘드리아 및 가용성분획을 분리하고 diamine oxidase의 활성도를 관찰하였으며, 기질에 대한 특이성과 기질에 대한 Km값을 측정하여 다음과 같은 결과를 얻었다. 1. 양막과 융모막 조직 1g당 diamine oxidase의 활성도는 각각 46.95unit alc 47.38unit이었다. 2. 양막조직의 diamine oxidase의 특이활성도는 융모막조직의 특이활성도 보다 2배이상 높았다. 3. 양막조직의 diamine oxidase는 가용성분획, 미토콘드리아분획 및 핵분획에 존재하며 가용성분획에 약 79.2%, 미토콘드리아분획에 약 10.8%, 핵분획에 약 10%가 분포되어 있었다. 4. 융모막조직에는 가용성분획에 약 83.1%, 미토콘드리아분획에 약 9.8%, 핵분획에 약 7.1%의 diamine oxidase가 분포되어 있었다. 5. 양막조직내에 diamine oxidase의 기질에 대한 활성도는 putrescine이 36.95±0.93unit/g of wet tissue, cadaverine이 45.56±3.04unit/g of wet tissue, spermidine이 40.81±1.48unit/g of wet tissue 및 spermine이 41.29±0.02unit/g of wet tissue로 histamine에 대한 활성도가 가장 높았으며, putrescine에 대한 활성도가 가장 낮았다. 6. 융모막 조직내에 diamine oxidase의 기질에 대한 활성도는 spermine이 54.35±0.95unit/g of wet tissue, cadaverine이 45.50±0.62unit/g of wet tissue, putrecine이 37.41±0.44unit/g of wet tissue, spermidine이 35.64±0.62unit/g of wet tissue 및 histamine이 31.35±0.71unit/g of wet tissue순이며 histamine에 대한 활성도가 가장 낮았다. 7. 양막조직의 diamine oxidase의 spermine, histamine, cadaverine, spermidine 및 putrescine에 대한 Km값은 각각 0.526mM, 0.725mM, 0.990mM, 1.250mM 및 1.316mM이었다. 8. 융모막조직의 diamine oxidase의 spermine, cadaverine, putrescine spermidine 및 histamine에 대한 Km값은 각각 0.676mM, 0.980mM, 1.282mM, 1.538mM 및 2.381mM이었다.

The activities of diamine oxidase in cytoplasm, mitochondria and nuclear fractions isolated from human amniotic and chorionic tissues were measured together with some biochemical characteristics and Km values to substrates. The results were summarized as follows: 1. Diamine oxidase activities in amniotic and chorionic tissues were 46.95 and 47.38 units/g of wet tissue, respectively. 2. The specific activity of diamine oxidase in amniotic tissue was twice as high as that in chorionic tissue(8.61 units/mg protein: 3.46 units/mg protein). 3. Diamine oxidase was distributed 79.2% in cytoplasm, 10.8% in mitochondria and 10.0% in nuclear fractions of amniotic tissue. 4. Diamine oxidase was distributed 83.1% in cytoplasm, 9.8% in mitochondria and 7.1% in nuclear fractions of chorionic tissue. 5. Activity of diamine oxidase in amniotic tissue was different according to substrates: 36.05±0.93 units/g of wet tissue in putrescine, 45.56±3.04 units/g of wet tissue in cadaverine, 63.58±3.34 units/g of wet tissue in histamine, 40.81±1.48 units/g of wet tissue in spermidine and 41.29±0.02 units/f of wet tissue in spermine. The activity was the highest in histamine and was the lowest inputrescine. 6. Activity of diamine oxidase in chorionic tissue was different according to substrates: 54.35±0.95 units/g of wet tissue in spermine, 45.50±0.62 units/g of wet tissue in cadaverine, 37.41±0.44 units/g of wet tissue in putrescine, 35.64±0.62 units/g of wet tissue in spermidine and 31.35±0.71 units/g of wet tissue in histamine. The activity was the lowest in histamine. 7. Km value of dimine oxidase in amniotic tissue was 0.52mM in spermine, 0.725mM in histamine, 0.990mM in cadaverine, 1.250mM in spermidine and 1.316mM in putrescine. 8. Km value of diamine oxidase in chorionic tissue was 0.676mM in spermine, 0.980mM in cadaverine, 1.282mM in putrescine, 1.538mM in spermidine and 2.381mM in histamine.
