시험관내 태반기관 배양에서 LHRH가 hCG 및 Progesterone 분비에 미치는 영향

저자들은 태반조직의 b-hCG및 progesterone분비 조절기능에 대한 LHRH역할을 알기 위하여 23예의 경상 산모로부터 만출된 태반조직을 이용한 시험관내 기관 배양법으로 LHRH를 투여하지 않은 태반조직(대조군)과 LHRH를 투여한 태반조직(실험군)으로부터 유리된 b-hCG 및 progesterone을 방사면역 측정법으로 측정하고 또 전자현미경에 의한 미세구조의 형태학적 변화를 관찰함으로서 다음과 같은 결론을 얻었다. 1. b-hCG값은 대조군에서 배양 제 1일과 2일에 각각 259+-23mIU/100mg, 102+-20mIU/100mg이상으로 증가하였으며(p<0.01) 배양일이 경과함에 따라 점차 감소하였으나, 배양 제 6일과 7일에는 87+-20bmIU/100mg, 88+-10mIU/100mg이상으로 재상승하였다. 또 LHRH의 용량이 증가함에 따라 b-hCG값은 용량-반응관계에 비례하였다. 3. Progesterone값은 대조군에서 배양 제 1일과 2일에 각각 187+-40ng/100mg이었으며, 배양일이 경과함에 따라 점차 감소하였다. 4. Progesterone값은 실험군의 전예에서 배양 제 1일과 2일에 각각 212-31ng/100mg, 102+-19ng/100mg이상이었으나 배양일이 경과함에 따라 점차 감소하였으며, 재상승을 보여주지 아니하였고, LHRH용량에 따른 변동도 관찰할 수 없었다. 5. 전자현미경 소견에서 실험군에서는 대조군에 비하여 다수의 소포, polyribcsome 및 내형질망이 관찰되었으며 잘발달된 golgi체의 지방적이 관찰되었다. 이상의 결과로 보아 LHRH는 태반조직의 b-hCG분비조절기능에 관여하는 것으로 생각되며, progesterone분비에는 직접적으로 관여하지 않는 것으로 생각되나 추후 더 많은 연구가 필요하다고 사료된다.

To evaluate the role of LHRH on b-hCG and progesterone production by human placental tissue, authors studied on placental tissues expelled from 23 normal term pregnant women by using In vitro organ culture. Authors measured b-hCG and progesterone secreted from placental tissue without (control group) and with administration of LHRH(experimental group) by using radioimmunoassay and observed morphological change of microstructure by electromicroscope. The results were as follows: 1. In control group, the values of b-hCG on the first and the second day of culture were 259+-23 and 102+-29mIU/100mg respectively and were gradually decreased with days of culture. 2. In experimental group, the values of b-hCG on the first and the second day of culture in all the cases were increased over 321+-42 and 137+-16 mIU/100mg respectively(P<0.01), and were gradually decreased with days of culture. However, the values were again increased over 87+-20 and 88+-10mIU/100mg on the 6th and the 7th day of culture respectively(P<0.01). and the values of b-hCG was proportional to dose-dependent response by increasing amount of LHRH. 3. In control group, the values of progesterone on the first and the second day of culture were 187+-40 and 85+-25 ng/100mg respectively and were gradually decreased with days of culture. 4. In experimental group, the values of progesterone on the first and the second day of culture were over 212+-31 and 102+-19ng/100mg respectively and were gradually decreased with days of culture. However, neither reincrease nor change according to amount of LHRH was seen. 5. In observation by electromicroscope, a large number of vesicle, polyribosome, endoplasmic reticulum, well-developed Golgi complex and fat droplet was seen in experimental group compared with control group. From the above results, it is thought that LHRH is concerned in production of b-hCG by placental tissue and is not concerned directly in production of progesterone which is required more studies afterwards.
