The class of polymorphic loci, known as VNTR(variation number of tandem repeat), are highly polymorphic in humans. In the present study, allele and genotype frequencies of the Human β-actin related pseudogene(ACTPB2) and Apolipoprotein B(ApoB) gene were determined among Koreans by using amplified fragment length polymorphism (Amp-FLP) technique. In ACTPB2 gene, 23 alleles and 36 genotypes were observed, whereas 11 alleles and 22 genotypes were determined in ApoB 3' gene among over 100 unrelated Koreans. In ACTPB2 and ApoB 3' genes, heterozygosity (H) was 0.817 and 0.706, and the power of exclusion (POE) was 0.956 and 0.850, respectively. The probability of sharing the same DNA band was 2.75×10-2 for ACTPB2 and 14.7×10-2 for ApoB 3'. Mendelian inheritance for the alleles at the two VNTR markers could be demonstrated in 20 paternity cases. The mutation rate of ACTPB2 gene was 0.04, however, the mutated band was not found in ApoB 3' gene. The data presented here on allele frequencies and mutation rates provide preliminary data supporting validity of ACTPB2 and ApoB 3' genes in paternity analysis among Koreans.
