SPP2 gene product of yeast Saccharomyces cerevisiae is involved in pre-mRNA splicing. To further characterize the role of SPP2 gene product in pre-mRNA splicing, we have determined base sequences of 6 ts spp2 alleles, M1 to M6, which had previously been isolated. The M2 and M3 had a point mutation at the 106th codon, which introduced a substitution of glycine with glutamic acid. The M4 and M6 had the same mutation as that of M2 and M3, although they had several other silent mutations. Substitutions at 4 and 2 amino acid residues were found in the M1 and M5, respectively. Upon computer analysis, no significant difference in secondary structure and hydropathy profile was observed between wild type SPP2 and a point mutation-containing M2 or M3. All the ts strains containing M1 to M6 showed a slight splicing defect even at a permissive temperature, but they all showed a significant splicing defect at a nonpermissive temperature.
