DNA Helical Periodicity Imaged by Scanning Tuneling Microscopy

pBR322 DNA molecules were deposited on highly oriented pyrolytic graphite and imaged in air with the scanning tunneling microscope. Clear view of the helicity of the uncoated DNA was evident. The pitch periodicity ranged from 38 Å to 47 Å, and from 42 Å to 50 Å, and the diameter ranged from 20 Å to 25 Å. Minor as well as major grooves were distinct from the vertical displacement profile. A 3 dimensional image clearly revealed the side view of the protruding sugar phosphate backbones. A transmission electron microscope picture of pBR322 DNA is also presented to compare the quality of images and the scales of viewing area.
