An actin gene from red pepper was cloned. Polyadenylated RNA purified from red pepper seedling was assayed for the actin mRNA by Northern hybridization with the Dictyosterium discoideum actin cDNA clone as a probe. Double-stranded cDNA synthesized from the polyadenylated RNA was cloned in EcoRV site of pBR322, and the cDNA bank was screened with the actin cDNA probe of D. discoideum. The putative clone was characterized by Northern hybridizations. The clone was designated as pRPAcI. We determined the nucleotide sequence of pRPAcI for the carboxy terminal half of the protein. The deduced amino acid sequence has high homology with other plant actins.
