Molecular Cloning of a rec - like Gene from Schizosaccharomyces pombe

We have constructed a Schizosaccharomyces pombe expression genomic library in λgt11 bacteriophage vector and isolated a rec-like gene by immunological screening with E. coli recA antibody. Forty putative clones were isolated from the first screening, 36 from the second, and 16 from the final. The size of the inserts ranged from 3.5 to 4.3 kb. Among these clones, one clone harboring 4.3 kb insert was selected and the insert DNA was introduced into Bluscript SK vector. The constructed plasmid was designated to pLAP30 and mapped with various restriction enzymes. The E. coli XL1-Blue transformed with the pLAP30 plasmid produced a β-gal/rec-like fusion protein. To verify that the fusion protein is composed of β-gal and rec-like proteins, Western blot analysis was performed with both antibodies raised against recA and β-gal proteins, respectively. The fusion protein was found to be recognized by both antibodies, indicating that the fusion protein consists of β-gal and rec-like proteins.
