We have identified and partially purified a protein in mitotic Schizosaccharomyces pombe cells by polyclonal antibodies raised against Escherichia coli RecA protein. By Western blotting on SDS-PAGE a major 74 kDa protein was detected in whole cell extracts which was prepared with high salt extraction and concentrated by addition of 0.48g of solid ammonium sulfate per milliliter. Four protein bands of 74 kDa, 62 kDa, 42 kDa, and 40 kDa were recognized in DEAE-cellulose chromatography which were eluted by 0.2 to 0.5 M NaCl linear gradient. Of which the DEAE-cellulose eluents, active fractions were pooled and analysed by denatured DNA cellulose chromatography. The protein also seems to be slightly induced by UV-irradiation or MMS treatment. These results suggest that a regulatory network which is controlled by Rec-like protein must be existed in Schizosaccharomyces pombe.
