Identification and Analysis of tRNA from Aspergillus nidulans by Partial Enzymatic Cleavage

Total tRNAs of A. nidulans were extracted by hot phenol extraction and purified with DEAE cellulose chromatography. The specific tRNAs were separated on the two-dimensional polyacrylamide gel. tRNA from single spot was eluted and labeled with r-^(32) P-ATP. After partial digestion with RNases (T₁, U₂, Phy L, B. cereus, CL 3), each sample was loaded on the sequencing gel. From the sequencing data, the testing tRNA was identified as tRNA^(Arg)_(ACG). tRNA^(Arg)_(ACG) shows considerably more homology with yeast (60% homology) than with prokaryotic (E. coli; 30% homology). The invariable sequences were observed. Further sequence analysis is going on by TLC method for the identification of modified bases.
