Avian reovirus (ARV) is the pathogenic agent of tenosynovitis and malabsorption syndrome in broiler, which has caused significant economical losses due to poor feeder efficiency and stunting. In order to determine the prevalence of ARV infection in poultry farms in Jeonbuk province, Korea, we performed a surveillance study by testing 179 cecal samples from 131 broiler farms for virus detection, and 1,181 serum samples from 33 broiler farms (n=292) and 22 broiler breeder farms (n=1,525) for antibody detection in the province. Virological examination using RT-PCR showed that ARV were detected in 26.0% of the tested farms (34/131),with the highest positive rates in broilers of 6 days old or more in summer season. In serological test using ELISA, broiler and broiler breeder farms examined were all ARV antibody positive. In broiler, the positive rate and antibody titers showed a tendency to decrease with age in contrast to those of broiler breeders. Our results indicate that ARV is ubiquitous in broilers and broiler breeders in the province.

Naegleria fowleri, a pathogenic free-living amoeba, leads to a fatal infection known as primary amebic meningoencephalitis (PAM) in human and animals. PAM is an acute, fulminant, necrotizing, and hemorrhagic disease that leads to death in approximately seven days. In this study, we investigate the cytotoxicity of target cells and the secreted molecules of N. fowleri under the non-contact condition. The target cell (U87MG cell) treated with N. fowleri lysates showed no morphological changes and no cytotoxicity. By contrast, the U87MG cells co-cultured with N. fowleri trophozoites under the non-contact condition induced morphological changes and reduction in number. When U87MG cells were co-cultured with N. fowleri trophozoites under the non-contact condition for 30 min, 2 hr, and 4 hr, the levels of cytotoxicity of target cells were 32.3, 35.5, and 37.8%, respectively. Particularly, when the ratio of amoeba to target cells is 10 to 1, the level of cytotoxicity of target cells was 49.7% at 30 min. To show the proteins secreted from N. fowleri under the non-contact condition, we carried out 2D electrophoresis and observed 6 major proteins. Finally, these results suggest that the molecules released from N. fowleri under the non-contact condition induce the cell death and this process is an important step in pathogenesis of N. fowleri.

Q fever is a zoonosis caused by Coxiella burnetti that is important not only for livestock but also for humans. This research aims to investigate infection rate of Q fever by examining the prevalence of antibody titers in cattle in Sejong City. From March to October 2018, 505 samples of Hanwoo serum and 47 samples of bulk-tank milk were collected and analyzed using ELISA. According to the result, 25 (53.2%) out of 47 bulk-tank milk samples were antibody positive and 28 (5.5%) out of 505 cows were antibody positive. The seroprevalence of C. burnetii increased with age (<1 y=0%, 1∼2 y=4.1%, 3∼5 y=7.5%, ≥6 y=9.6%) and was higher in females (6.5%) compared to males (0%).

Claw lesion is one of the major causes of lameness in sows and the lameness is one of the reasons for culling of sows from the swine herd. This survey was conducted on sows in total of 8 farms with a total of 684 pigs. The lesions observed in the study were horizontal and vertical claw cracks (CWH & CWV), differences in the length of toes (T) and/or in the length of dew claws (DC) according to crates, breeds and parity of sows. Scores of claw lesions were assigned on 0, 1, 2, and 3 at each foot depending on the severity of the claw lesions. The sows' parity was classified into 1 to 2, 3 to 4, 5 to 8. Prevalence rate of claw lesions in sows raised on punched plastic crate was higher than that in sows raised on tribar crate. In prevlence rate by breeds, inbreed Landrace sows showed higher than that of Yorkshire sows. According to 4 claw lesions, the length of DC was the highest at parity 3∼4 and CWV was the lowest at parity 3∼4 in F1 crossbred sows raised on punched plastic crates, respectively. In Yorkshire breeds raised on punched plastic crates, the length of DC at parity 5∼8 was the lowest and CWH was the highest at parity 3∼4, respectively. In Landrace breeds raised on tribar crates, CWH was the highest at parity 1∼2 and CWV was the lowest at parity 3∼4, respectively. The high parity in F1 crossbred sows showed high prevalence rate of claw lesions than that of low parity and other inbreed sows, respectively. These results may be useful as elementary data in establishment of welfare quality protocol and preventive measures to reduce economic losses from craw lesions in sows.

Blood test is a useful tool in establishing medical treatment for livestock. It provides information such as disease diagnosis, treatment effects, prognostic judgment, and health status. This study compared the value of erythrocytes and leukocytes among conventional, transgenic miniature, and transgenic conventional pigs aged six months to 24 months. Further, it analyzed the aspects of hematological value changes according to the pigs' ages. As a result, the number of red blood cells (RBC), which include hemoglobin, and hematocrit, and the number of white blood cells (WBC), which include neutrophils, and lymphocyte, were high among transgenic miniature pigs, compared with the conventional and transgenic conventional pigs. Conventional pigs showed similar values of RBC and WBC regardless of transgenesis. In comparing their age, the RBC decreased as the age increased compared with the pigs among all the three groups aged of 6∼12 months. On the other hand, WBC and neutrophils showed no significant difference regardless of different ages among all the three groups. However, various counts in RBC and WBC were mostly found to be higher in each age in transgenic miniature pigs than in conventional and transgenic conventional pigs. The results of this study show that the values of RBC and WBC were generally higher in transgenic miniature pigs than in conventional and transgenic conventional pigs. Based on this research, hematological values can be widely used in diagnosing diseases or checking the health status of transgenic pigs that are used as disease models, organ transplant source and alike.

Porcine reproductive and respiratory syndrome (PRRS) is characterized by reproductive failure in sows and respiratory distress in all age pigs. Porcine respiratory disease complex (PRDC) is a disease caused by opportunistic bacterial infection secondary to a weakened immune system by a preceding respiratory infection. In this study, we tried to compare the immune responses in PRRS and PRDC groups to clearly characterize the disease severity. Eighty-five pigs were infected with various Korean field PRRS virus strains. Infected animals were classified into PRRS (n=32) and PRDC (n=53) groups based on lung lesions such as interstitial pneumonia, suppurative pneumonia, and pleuropneumonia. The immune cell population of bronchoalveolar lavage cells (BALc) was evaluated on 14 and 28 days post infection (dpi) and PMBC cytokine expression was measured on 0, 3, 7, 14 dpi to investigate early inflammatory reactions. Pulmonary lesion severity was negatively correlated with alveolar macrophage (AM) in both PRRS and PRDC groups on 14 and 28 dpi. AM in BALc was less populated in PRDC group on 28 dpi compared to PRRS group. AM in BALc was significantly less populated in PRDC group on 28 dpi compared to 14 dpi. In addition, cytotoxic T lymphocyte (CTL) in BALc was higher populated in PRDC group on 14 dpi and 28 dpi compared to PRRS group. In the case of PBMC cytokine TNF-α, IFN-α, IL-1β, IFN-γ, FoxP3, and IL-2, the PRRS group showed higher expression than the PRDC group on 7 dpi, 14 dpi, 7 dpi, 14 dpi, 14 dpi, and 14 dpi, respectively. On the other hand, in the case of IFN-β, IL-6, IL-8, IL-4, and IL-17, the PRDC group showed higher PBMC cytokine expression at 14 dpi, 7 dpi, 14 dpi, 3 dpi, and 3 dpi, respectively, than the PRRS group. Based on these results, our study could characterize differential immune responses in pigs with PRRS or PRDC.

To detect Single nucleotide polymorphisms (SNP) markers associated with Bovine viral diarrhea virus (BVDV) and Bovine respiratory syncytial virus (BRSV) S/P ratio in Korean Holstein dairy cattle, Genomewide association study (GWAS) was performed using Illumina BovineSNP50 Beadchip. The number of phenotype data and genotype data were 107, and 294. respectively. Phenotype data were collected for four periods (0 week, 1 week, 4 week, 24 week) after having vaccinated (0 week no vaccinated period). A total of 36,257 SNPs was remained after quality control had been done by PLINK. The result of GWAS showed 6 SNP markers (BTB-01704243, BTB-01594395, ARS-BFGL-NGS-118070, ARS-BFGL-NGS- 111365, BTA-65410-no-rs, Hapmap38331-BTA-61256) under BVDV and 4 SNP markers (ARS-BFGLNGS- 109861, Hapmap53701-rs29017064, ARS-BFGL-NGS-71055, BTA-11232-no-rs) under BRSV. And also, 10 candidate genes found through 10 SNP markers (TBX18, CEP162, PAFAH1B1, METTL16, BRCA1, RND2, POLK, ENSBTAG00000051724, ADAM18, NRG3).

The complete blood count and serum biochemical parameters are essential tools for diagnosis and monitoring of physiological and disease conditions in dogs. These parameters may be affected by the breed and age of dogs. This study aimed to compare the changes of complete blood count and serum biochemical parameters according to the increasing age in 156 Sapsarees. In this study, the Sapsarees were clustered into ten age groups: four to six months, seven to nine months, ten to twelve months, one year, two years, three to four years, five to six years, seven to eight years, nine to ten years, and, eleven to twelve years old. In the result of the complete blood count, the total red blood cell count (P<0.001), hematocrit (P< 0.001), and hemoglobin (P<0.001) values were significantly increased from 4∼6 months to 2 years old Sapsarees. While the mean corpuscular volume (P<0.001) and mean corpuscular hemoglobin (P< 0.001) values were significantly increased from 4∼6 months to 3∼4 years old. The mean corpuscular hemoglobin concentration was significantly (P=0.037) increased from 4∼6 to 10∼12-month-old Sapsarees. Total white cell count was significantly (P<0.001) decreased from 1 year to 9∼10 years old. The result of differential white blood cell count showed that neutrophil count was significantly (P< 0.001) increased from the age 4∼6 months to 11∼12 years old Sapsarees, whereas, lymphocyte (P< 0.001), monocyte (P<0.001), and eosinophil (P=0.042) counts were significantly decreased from 7∼9 months to 7∼8 years, 4∼6 months to 9∼10 years, 3∼4 to 5∼6 years old, respectively. In the serum biochemistry result, the creatinine concentration was significantly (P<0.001) increased from 4∼6 to 10∼ 12 months, but significantly (P=0.006) decreased from 7∼8 to 10∼12 years old. Phosphate concentration was significantly (P<0.001) decreased from 3∼4 to 9∼10 years old, but significantly (P=0.021) increased from 9∼10 to 11∼12 years old. Calcium concentration significantly (P<0.001) decreased from 10∼12 months to 11∼12 years old. Total protein concentration significantly (P<0.001) increased from 4∼6 months to 2 years old. While concentrations of albumin (P=0.004) and globulin (P<0.001) was significantly increased from 4∼6 months to 3∼4 years old, and from 4∼6 months to 11∼12 years old, respectively. The alkaline phosphate concentration was significantly (P<0.001) decreased from 4∼6 months to 2 years old. We found that there are differences in the hematological parameters in relation with the increasing age in Sapsaree breed.

9Sequence analysis of the fusion protein gene of Newcastle disease virus isolated from breeder ducks in Korea

Newcastle disease (ND) is an infectious poultry disease that caused high mortality and reduced egg production. NDVs are regularly present in the domestic duck population. And ducks play a possible role in the maintenance and transmission of NDVs. While we were monitoring the Avian Influenza, NDVs were isolated from field samples by accident. So we analysed the biological and genetic characteristics of these viruses. Lentogenic NDVs were isolated from two farms among twenty breeder duck farms. The ages of ducks were 39 weeks old in the 'A' farm and 3∼72 weeks old in the 'B' farm. And they were not inoculated with the NDVs vaccine. In the biological characteristics, the both viruses which separated from the farm 'A' and 'B' were thermostable. The amino acid sequence of a site from 112 to 119 in the fusion (F) protein was 'GKQGRLIG' which has monobasic motif in the samples of both farms. And this means the separated NDVs are lentogenic. Phylogenetic analysis was performed by entire nucleotide sequence of F protein. The virus strains from the A farm (MN095239) and the B farm (MN095240) belonged to class II genotype I. Using the analysis of whole F protein nucleic acid sequence, the MN095239 (GenBank) had homology with Ulster strain about 99.95% and the MN095239 (GenBank) had homology with KR/CK/KU_LBM255/09 strain about 99.89%. NDV surveillance is needed to investigate epidemiological relationship of domestic breeder duck isolates in Korea.

10Multidrug resistance of coagulase-negative staphylococci isolated from rescued wild animals

Wildlife is a bio-indicator of environmental pollution by antimicrobial resistant bacteria or genes, however, there is no information on antimicrobial resistance in wildlife-origin bacteria. This study aimed to investigate the normal microbiota of staphylococci and their antimicrobial resistance in wildlife that did not take any antimicrobials. After sampling and bacterial isolation/identification, antimicrobial resistance profiles were examined by broth microdilution test, Kirby-Bauer disc diffusion test and mecA genetargeted PCR. Of 90 isolates from wildlife, 83 were coagulase-negative staphylococci while only 7 were coagulase-positive staphylococci. Methicillin-resistance was found in 63 (70%) isolates and 35 of 90 (38.9%) isolates were multidrug-resistant staphylococci. When considering that all of the animals did not take any medication or contacted any medical device before the sampling, the results indicate significantly high prevalence of antimicrobial resistance in wild environments. Further study would be necessary to investigate the transmission route of antimicrobial resistance.
