Glutamate acts as an important neurotransmitter in brain. However, high concentration of glutamate showed an excitatory neurotoxicity and resulted to neuronal cell death. Neuronal cell death is known for one of the reason of Alzheimer’s disease, a neurodegenerative disease. We tried to find neuroprotective medicinal plants by neuroprotection activity against glutamate injured HT22 cells as a model system. In the course of bioscreening of various medicinal plants, Taraxacum platycarpum extract showed significant neuroprotective activity. We tried to elucidate mechanisms of neuroprotective activity. T. platycarpum extract reduced ROS and intracellular Ca2+ concentration increased by glutamate induced neurotoxicity. In addition, mitochondrial membrane potential was restored to the control level. Also, glutathione level, glutathione reductase and glutathione peroxidase activity were increased by T. platycarpum extract treatment. These data suggested that T. platycarpum showed neuroprotective activity via antioxidative activity.

Adenophora stricta Miq. (Campanulaceae) is an annual herb, which has been used as a traditional medicine in Korea, Japan and China to treat bronchial asthma, tonsillitis, and hypertension. In this study, 12 compounds were isolated from the roots of A. stricta and isolates were identified to be methyl adenophorate (1), decursidin (2), L-tryptophan (3), D-1,2,3,4-tetrahydronorharmane-3-carboxylic acid (4), vanillic acid 4-O-β-D-glucopyranoside (5), syringic acid 4-O-β-D-glucopyranoside (6), vanillin (7), vanillic acid (8), p-hydroxybenzaldehyde (9), p-hydroxybenzoic acid (10), p-hydroxyacetophenone (11) and linoleic acid (12). Decursidin (2) and D-1,2,3,4-tetrahydronorharmane-3-carboxylic acid (4) is firstly reported from A. stricta in current study.

An endophytic fungus, Penicillium sp. JVF17, was isolated from a leaf of Vitex rotundifolia in coastal area of Jeju island. Chemical investigation of this fungal strain resulted in the isolation of four compounds, piceol (1), cyclo (L-Pro-L-Val) (2), isochromophilone VI (3), and dicitrinin A (4). Their chemical structures were elucidated by comparison of their spectral data such as NMR and ESIMS with reported literature values.

In the present study, the effect of Z. jujuba Miller var. inermis Rehder extracts on the secretion of atopic dermatitis (AD)-related cytokines and hyaluronidase activity was investigated. We prepared four fractions, butylene glycol (JB), ethanol (JE), and water (JW), with Z. jujuba Miller var. inermis extracts. JW significantly reduced the secretion of interleukin-8 and JE reduced the secretion of tumor necrosis factor-alpha in human keratinocyte HaCaT cells. Also, hyaluronidase activity was measured by enzyme assay and the fractions inhibited the activity in a dose-dependent manner. In addition, the human dermal fibroblast, HDF-n cells were treated with the extracts and antioxidant activities were measured. The results showed that the extracts increased the free radical scavenging activity and the superoxide dismutase activity. Taken together, Z. jujuba Miller var. inermis extracts reduced the secretion of AD-related cytokines and inhibited the hyaluronidase. In addition, the extracts showed antioxidant activity.

Methanol extract of Helianthus tuberosus L. (Compositae) flower was investigated to research the anti-oxidative activity by using a Caenorhabditis elegans model system. Ethyl acetate soluble fraction of the methanol extract showed the most potent DPPH radical scavenging activity. The ethyl acetate fraction was measured on its activities of superoxide dismutase (SOD), catalase, and oxidative stress tolerance with reactive oxygen species (ROS) level in C. elegans. Furthermore, in order to verify if regulation of stress-response gene is responsible for the increased stress tolerance of C. elegans which treated by the ethyl acetate fraction, we checked SOD-3 expression using a transgenic strain. Consequently, the ethyl acetate fraction of H. tuberosus flower increased the catalase and SOD activities in a dose-dependent manner in C. elegans, reduced ROS accumulation dose-dependently. Besides, the ethyl acetate fraction-treated CF1553 worms showed higher SOD-3::GFP intensity than the control group.

Inflammation that is considered to be mainly related to pathogenesis of atopic dermatitis (AD) is the biological response of a host to stimuli, such as cellular injury or infection. In this study, we investigated the anti-inflammatory and antioxidative activities of white ginseng roots by ultra high pressure extraction (Gin-UHP), fermentation followed by ultra high pressure extraction (Gin-UHPF), and polyol extraction (Gin-POL). As a result, ginseng extracts were able to decrease the secretion of pro-inflammatory cytokines (interleukin-8 and tumor necrosis factor-alpha) and immunoglobulin E. Also, Gin-POL had the highest DPPH radical scavenging activity and when we compared the SOD-like activity, Gin-UHP had the highest. Moreover, we looked into the effect of these ginseng extracts on anti-aging to show the possible usefulness as a raw material of cosmetics. As a result, ginseng extracts were able to reduce the production of melanin, and inhibit the tyrosinase and elastase activities in a dose-dependent manner. The extracts also decreased the expression of MMP-1 and had a significant hyaluronidase inhibitory activity. Taken together, these results demonstrate that ginseng extracts may have an improvement effect on AD by using its anti-inflammatory and antioxidant properties.

In this study, extracts of bark from Ulmus davidiana var. japonica and its fractions were investigated the antioxidative, antibacterial and tyrosinase inhibition activity for physiological activity towards functional applications. In the measurement of DPPH radical scavenging activity, the ethyl acetate fraction showed higher radical scavenging ability than others. Moreover, in the tyrosinase inhibition assay, showed that the ethyl acetate fraction has good inhibition effects. Results of the DPPH radical scavenging and tyrosinase inhibition activity are related with the total polyphenol concentrations of ethyl acetate fraction. In antibacterial activity used to find out by utilizing the disc diffusion assay, chloroform fraction showed strong effect against Staphylococcus aureus and S. epidermidis. These results are related with the flavonoid contents of chloroform fraction. On the other hand, in the L929 cell viability measurement by MTT assay, the hexane, butanol and aqueous fraction treated at high concentration were showed cytotoxicity. But the others samples were exhibited a moderate viabilities. As a result of investigated the antioxidant and tyrosinase inhibition activity, the ethyl acetate fraction could be applicable for cosmetics related fields. And the chloroform fraction showed significant antibacterial activity against S. aureus and S. epidermidis.

This study compared the contents of phenolic components depending on the extracting conditions of Acanthopanax koreanum stem to provide basic information for developing Acanthopanax koreanum-based functional foods. Our findings show that the content of total phenolic component peaked at 16 hours of extraction (WAK-16, 7.22%) and when water extracted at 100℃. However, 11-hours water extraction (WAK-11) showed highest eleutheroside B concentration of 1.32%, a main component of A. koreanum, and the level of chlorogenic acid concentration was the highest when 1-hour water extraction (WAK-1) was conducted, being 2.12%. Moreover, highest concentration of eleutheroside E was observed in 16-hours water extraction (WAK-16) as 1.49%. With 60-hours water extraction (WAK-60), the content of syringaresinol, an active phenolic aglycon substance, concentration was the highest with the value of 0.10%. Isofraxidin showed the highest concentration of water extract(0.09%) for 20 hours (WAK-20) and 0.42% for sinapyl alcohol[16 hours (WAK-16)].

In our previous study, we found uracil, adenosine, protocatechuic acid, syringin (eleutheroside B) and scoparone (6, 7-dimethoxycoumarin) in the Oplopanax elatus Nakai Stem. High-performance liquid chromatography (HPLC) -UV was used to quality and quantify the internal marker compounds in the O. elatus extract after validation of method with linearity, limit of detection (LOD), limit of quantitation (LOQ), accuracy and precision. The specificity assessment visually confirmed that the substance was detected without the introduction of other substances. The established method showed high linearity of the calibration curve and coefficient of correlation (R2) of over the 0.999. HPLC was reported as five standard compounds equivalent using the following linear equation based on the calibration curve. The accuracy of measurement was 84.34 ~ 119.74% and the relative standard deviation (RSD) value was 0.28 ~ 1.60%. In addition, our established method showed high repeatability. The RSD value was 1.10 ~ 6.81%. So, we found the amount of the internal marker compounds in the O. elatus extract. These results demonstrated that can be used to quality evaluation of the O. elatus.

The aim of this study was to investigate the potential of Acanthopanax sieboldianum (Makino) Koidz (ACS) as a potent antioxidant and antidiabetic agent. The antioxidative and alpha-glucosidase inhibitory activities were examined using the methanol extracts and solvent fractions from ACS-leaf and ACS-stem. Antioxidative activities were measured by in vitro methods such as DPPH and ABTS radical scavenging activity and superoxide dismutase (SOD) activity. When the chloroform and ethyl acetate fractions of ACS-leaf and ethyl acetate fractions of ACS-stem were compared with the control, the SOD-like activity was impaired even at the low treatment concentrations. In addition, the ethyl acetate fractions of ACS-leaf and ACS-stem showed alpha-glucosidase inhibition activities at low treatment concentrations. Analysis of the major components in the fractions of ACS-leaf and ACS-stem was also performed using HPLC. Finally, astragalin, isoqurecetin, chlorogenic acid and caffeic acid contents were measured. Based on this work, we propose that ACS-leaf and ACS-stem have great potential as natural antioxidant and antidiabetic materials related to health benefits.
