The spatial distribution and abundance of fish species was estimate near Dok-do, East Sea of Korea by conducting midwater trawl survey with hydroacoustic method. Acoustic and midwater trawl data were collected onboard the R/V Tamgu 21 during 7-8 March 2016. Acoustic data were collected at the frequencies of 38 and 120 kHz and converted to nautical area scattering strength (m2/nmile2). Species are distributed mainly over an extent of 30-55 m, generally around southwest of Dok-do; the squid Watasenia scintillans was the dominant species found. The length-weight function and target strength of squid and other fishes were used to estimate the W. scintillans stock at 2499.1 tons and other fish at 43.8 tons during the winter season. The hydroacoustic method offers an approach for the determination of the biomass and vertical and spatial distributions of species aggregating near Dok-do, East Sea of Korea.

Oysters Crassostrea gigas are a globally popular shellfish for human consumption. As filter-feeding bivalve mollusks, oysters may harbor many microorganisms and chemicals that could pose potential human health risks. The objective of this study was to investigate the suitability of cultured oysters for raw consumption or use in seafood products by measuring concentrations of harmful microorganisms and chemicals in their flesh. Microbial concentrations in cultured oysters were found to be: 1.0×102-6.0×104 CFU/g (viable cell counts), not detected (ND)-5.4×103 CFU/g (coliform bacteria), ND-1.3×102 CFU/g (E. coli), and ND-4.6×103 CFU/g (Vibrio parahaemolyticus). Other patho-genic bacteria, including Enterohemorrhagic E. coli (EHEC), Listeria monocytogenes, Staphylococcus aureus, and Salmonella spp., were not detected in any samples. Heavy metal concentrations of cultured oysters were ND-0.239 mg/kg (total mercury), ND-1.091 mg/kg (lead), ND-0.968 mg/kg (cadmium). The concentrations of benzo(a)pyrene ranged from 0.280-0.880 μg/kg. Paralytic shellfish poison ranged from ND-0.58 mg/kg, while diarrhetic shellfish poison was not detected. No radioactivity was detected. These results suggest that oysters intended for raw consump-tion or use in seafood products should be subjected to chemical and biological controls.

Sanitary evaluation of seawater and Pyropia sp. laver collected from the five major laver growing areas in Korea was performed four times over the course of a year. The seawater quality in four of these five areas was regarded as the clean area according to Korean criteria, but the seawater at one investigation site in Seoheon area was found to exceed the standard for fecal coliform. In the bacteriological safety analysis of laver (raw source), the percentages of samples not conforming to Chinese criteria at the five sites were 55.6% (Seocheon), 70.0% (Shinan), 81.8% [Jindo (Haenam)], 63.6% (Wando), and 28.6% [Goheung (Jangheung)]. Pathogenic bacteria were not detected in all laver samples. The food safety of laver (raw source) based on heavy metal concentration was confirmed using Korean criteria; the concentrations of heavy metals in laver samples collected from the major laver growing areas were 0.008-0.632 mg/kg wet weight (ww) lead, 0.024-0.137 mg/kg ww cadmium, 0.908-2.892 mg/kg ww total arsenic, and 0.003-0.013 mg/kg ww total mercury. Therefore, pollution source management and periodic monitoring of heavy metals may be required to improve the food safety of laver produced in these laver growing areas.

The object of this study is to assess physicochemical and microbiological hazards involved during the processing of shellfish products such as oysters, clam, and mussels. Samples including raw materials, intermediates, and final products in the processing process were collected from seven simple-processed shellfish facilities. In the samples obtained from the facilities, viable cell count and coliform group were detected less than that of the Korean Govern-ment guidelines. In addition, the high-risk bacterial pathogens such as Escherichia coli O157:H7, Salmonella sp., and Listeria monocytogenes were not identified in raw materials and final products. However, the low-risk pathogens, Staphylococcus aureus and Vibrio parahaemolyticus, were detected in some final products. The level of heavy metal content in the final products tested in this study meets the recommendations by the Korean Government standard guidelines. No foreign materials in the final products were also observed. Considering these results, it was concluded that no significant food hazards exists in the processing process of simple-processed shellfish products. However, it is essential to improve the food safety control in the shellfish processing facilities since S. aureus, a personal sanitary indicative bacterium, was continuously detected over seven simple-processed shellfish facilities.

To develop new functional materials, Saccharina japonica (S. japonica) was cultivated with Phellinus linteus (P. linteus) mycelia by solid culture. S. japonica was fermented with P. linteus mycelia. The various radical scavenging activities of extracts of fermented S. japonica with P. linteus mycelia (FSPM) were investigated and compared to the antioxidant capabilities of unfermented S. japonica and P. linteus mycelia. The antioxidant activities of FSPM extracts were evaluated using ferric reducing antioxidant power (FRAP) and 2,2`-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity. FSPM showed stronger free radical scavenging abilities than P. linteus mycelia or S. japonica alone. These results indicate that fermentation of S. japonica changes its chemical nature and could provide beneficial antioxidant effects.

Six experimental diets for juvenile sea cucumbers Apostichopus japonicus were evaluated and compared with current commercial diets. Sea cucumbers (50 per tank; initial mean weight 2.3±0.1g) in three replicates of seven groups were fed one of six experimental diets (ED1, 20% wild-harvested Sargassum thunbergii powder; ED2, 20% commercial S. thunbergii powder; ED3, 20% Undaria pinnatifida and Ascophyllum nodosum powders; ED4, 40% U. pinnatifida and A. nodosum powders; ED5, 10% brewer`s yeast; and ED6, 10% sea shell powder) or a commercial diet (CD) for 28 weeks. The survival rate in all groups was 85-94%, with no significant difference among the experimental groups. The mean body weight of the sea cucumbers was significantly different among the experimental groups 6 weeks after the feeding trial. The final mean weight of the sea cucumbers was highest in ED1, ED3, and ED4, followed by ED6, ED5, ED2, and CD, in decreasing order. The results of this study suggest that U. pinnatifida and A. nodosum powders could provide a substitute for the S. thunbergii powder commonly used in commercial sea cucumber diets. Thus, the EP4 diet formulation, which contains no S. thunbergii, could be used as a practical feed for juvenile sea cucumbers.

A feeding trial was conducted to evaluate the effects of partial replacement of fishmeal (FM) protein in a practical diet for rockfish Sebastes schlegeli juveniles with mealworm Tenebrio molitor meal (WM), in terms of growth per-formance, feed utilization, whole body composition and hematological parameters. Four isonitrogenous and isoen-ergetic diets were formulated to contain 8, 16, 24 and 32% WM (designated as WM8, WM16, WM24 and WM32). A FM-based diet without WM inclusion was used as a control. Triplicate groups of rockfish (2.6±0.07 g) were hand fed one of the experimental diets to visual satiation twice a day for 8 weeks. Weight gain of fish fed the WM32 diet was lower than that of control. No significant differences were found in daily feed intake, feed efficiency and protein efficiency ratio. The whole-body protein content of fish fed WM0 and WM8 diets were significantly higher than that of fish fed WM32 diet. No statistical differences were observed in hematological parameters. These results indicate that WM has potential as alternative to FM in practical diets for juvenile rockfish and can be used at an inclusion level of up to 24% without having a significant negative effects on growth and feed efficiency.

This study was performed to isolate bacteria that could control the nitrite levels in a biofloc technology (BFT) culture tank. Nitrite-eliminating bacteria were isolated from a BFT culture tank rearing goldfish, and the isolated bacterium exhibiting the most potent nitrite eliminating ability was labeled as the “NOBSB1” strain. Sequencing the 16S rRNA revealed that NOBSB1 is a species in the genera Bosea. NOBSB1 had the following characteristics with regard to nitrite removal: (1) it removed nitrite by functioning heterotrophically in the presence of a carbon source (sugars); (2) it eliminated nitrite most effectively within a temperature range of 20-30℃, but its activity decreased at temperatures above 35℃ and below 20℃; (3) it had optimum nitrite removal ability within a pH range of 6.0-8.0; (4) it removed nitrite more effectively under hypoxic than aerobic conditions. NOBSB1 inoculation did not decrease ammonia or nitrate levels, but eliminated nitrite in a BFT culture tank rearing common carp (Cyprinus carpio). After inoculating the NOBSB1 strain in a BFT culture tank, NOBSB1 controlled and sufficiently reduced the nitrite concentration in the tank.

We investigated the toxicity of zinc and aluminum hot-dip galvanized sheet steel to abalone Haliotis discus hannai via changes in the gill and hepatopancreas using histological and transmission electron microscopy analysis. Experi-mental groups were composed of one control and four exposure conditions (direct or indirect exposure to zinc and aluminum hot-dip galvanized sheet steel). In the control group, aluminum exposure groups (direct and indirect), and indirect zinc exposure group, abalone mortality was not observed until the end of the experiment, and no histopatho-logical changes were observed in the gill and hepatopancreas. However, the direct zinc exposure group exhibited 100% mortality. Ultrastructural analysis of the cytoplasm of ciliated and microvilli-bearing epithelial cells from gill filaments revealed electron-dense vesicles near the cell membrane and disruption of the nuclear membrane. We also observed swollen mitochondria and a loss of mitochondrial cristae. The hepatopancreas showed similar changes, and we detected highly electron-dense particles within the vesicles. These results suggest that abalone exposed directly to zinc hot-dip galvanized sheet steel experience acute toxicity, causing damage to cell organelles in the gill and hepa-topancreas and, finally, inducing mortality.

We observed the osteological development in larvae and juveniles of Korean spotted sleeper Odontobutis interrupta bred in the laboratory in April 2014. Immediately after hatching, the prelarvae, which were about 4.27 mm long, showed ossification of the premaxillary bones in the upper jaw and the dentary and articular bones in the lower jaw. At 7.11 mm, the larvae showed complete fusion of the post-cleithra and ossification of the scapulae with the appear-ance of one hole. At 8.65 mm, the larvae showed ossification of seven ribs from the third abdominal vertebra and an increase in the length of the neural spine and hemal spine. The number of caudal fin rays increased to 19. At 11.9 mm, the juveniles showed ossification of three procurrent rays on the side of the parhypural bone as well as ossification of two procurrent rays on the side of the epural bone, indicating the complete ossification of all spicules.

Vocalizations of the common dolphin Delphinus Delphis, were recorded from a single hydrophone while coastal visual survey was performed in the East Sea in March 2017. Like most Delphinidae, common dolphins produce whistles, echolocation clicks, and burst-pulsed calls, with repertoires that differ between species and geographically separated populations. This study focuses on using frequency analysis to classify sounds into three categories and to compare them with those of other common dolphin populations. The fundamental frequencies of the whistles were high in the 6 to 14 kHz range, while echolocation clicks were in the 40 to 90 kHz frequency band. These results are similar to high-energy distributions in the same frequency band in other regions. Based on this study of the acoustic characteristics of dolphins off the east coast of Korea, cetacean vocalizations can be classified using a database of cetacean sounds in Korea. In addition, this technique could improve data quality for visual whale surveys and could be applied to various other research subjects.
