Diabetes mellitus, commonly known as diabetes, is a group of metabolic disorders characterized by high blood sugar levels over a prolonged period. Diabetes has been found to show many acute complications such as cardiovascular disease, stroke, chronic kidney disease, foot ulcer and damage to eyes. Korean ginseng (Panax ginseng) has been traditionally known to normalize the functional deficiencies of the lung, spleen and stomach, and thus improve the secretion of body fluids, thereby quenching thirst, suggesting it to be effective in the treatment of diabetes. Experimental studies (in vitro and in vivo) have recently shown that Korean ginseng and its extracts exhibit antidiabetic effects, and also insulin secretion and sensitizing effects related to blood glucose control. Moreover, clinical trials on antidiabetic effects of Korean ginseng have been reported to show blood glucose control, improvement of insulin resistance, reduction of postprandial blood glucose level and improvement of serum lipids (TG, TC, LDL-C). These will be critically examined by means of in vitro studies, cell experiment, animal models and human trials with a focus on understanding of molecular mechanisms.

We previously reported that caffeic acid isolated from Lonicera japonica showed potent neuroprotective activities against glutamate injured neuronal cell death in primary cortical cells. In this study, we tried to confirm the neuroprotective activity in glutamate injured HT22 cells and elucidate mechanisms of neuroprotective action of caffeic acid. We used glutamate induced HT22 cell death as a bioassay system. The compound decreased reactive oxygen species increased by high concentration of glutamate treatment in HT22 cells. Also, Ca2+ concentration was decreased by this compound. This compound made mitochondrial membrane potential maintain to normal condition. This also affected anti-oxidative enzymes and glutathione contents. Treatment of this compound increased not only glutathione reductase and peroxidase to the control level and also amount of glutathione, an endogeneous antioxidant. These experimental results showed that caffeic acid isolated from L. japonica exerted potent neuroprotective activity through the anti-oxidative pathway.

The CHCl3 fraction of the MeOH extract of Ganoderma gibbosum (Ganodermataceae) exhibited cytotoxic activity on five cancer cell lines (MDA-MB-231, SK-hep1, A549, HCT116, and SNU638). Six highly oxygenated lanostane-type triterpenoids (lanostanoids) were isolated by column chromatography to test cytotoxicity on cancer cells. The five known lanostanoids were identified as gibbosic acids A, B, D, G, and H by comparison of molecular ion peaks with the literature data. The structure of a new lanostanoid, gibbosic acids I, was identified to be 3β,8β,15β,20S-tetrahydroxy-7,12,23-trioxolanost-9(11)-en-26-oic acid on the basis of NMR and MS spectroscopy. The three lanostanoids of gibbosic acids A, H, and I of the six isolates significantly suppressed the growth of cancer cells. In particular, the IC50 of gibbosic acid H was prominently low ranging from 2.64-6.56 μM.

This study was conducted to evaluate the effects of Padina arborescens and 1-O-myristoyl-2-O-oleoyl-3-O-(α- D-glucopyranosyl)-glycerol(MOGG), its active component, on the prevention of hair loss. The P. arborescens extract and MOGG inhibited the activity of 5α-reductase, which converts testosterone to dihydrotestosterone(DHT), a main cause of androgenetic alopecia. When immortalized rat vibrissa dermal papilla cells were treated with MOGG, the proliferation of dermal papilla cells significantly increased. In addition, we found that the P. arborescens extract and MOGG could open the KATP channel, which may contribute to increase hair growth. Furthermore, MOGG promoted PGE2 production in HaCaT cells. The results suggest that MOGG from the P. arborescens extract has the potential to treat alopecia via 5α-reductase inhibition, the proliferation of dermal papilla, the opening of the KATP channel and/or increase of PGE2 production.

The aim of this study was to investigate the epidermal moisturizing effects of artocarpin on normal human epidermal keratinocytes (NHEKs). To investigate the effects of artocarpin on NHEKs, cell viability and the expression of mRNAs related to skin hydration were measured. In addition, hyaluronic acid (HA)-ELISA assay was performed. Here, the effects of artocarpin on AQP3, HAS2, KRT1, and KRT10 mRNA expression, and on HA production, following UVA treatment were reported. The Quantitative real-time PCR results demonstrate that artocarpin increased AQP3, HAS2, KRT1, and KRT10 mRNA levels. The HA-ELISA assay revealed that artocarpin also increased HA production in NHEKs. Through these experiments, the epidermal moisturizing effects of artocarpin have been elucidated, providing evidence that artocarpin may be a potent cosmetic ingredient in skin anti-aging and moisturizing products. Based on these results, I anticipate that further research on the mechanisms of action of artocarpin may allow the development of not only cosmetics, but also medicines and healthcare foods.

In this study, 15 compounds were elucidated from the hot-water extract of Perillae Folium. Fifteen isolates were determined to be protocatechuic acid (1), caffeic acid (2), (R)-rosmarinic acid (3), (S)-shisoflavanone A (4), luteolin-7-O-β-Dglucuronopyranoside (5), scutellarein-7-O-β-D-glucuronopyranoside (6), apigenin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-Dglucuronopyranoside (7), luteolin-7-O-β-D-glucuronopyranosyl(1→2)-O-β-D-glucuronopyranoside (8), kelampayoside A (9), trans-N-feruloyloctopamine (10), 3-(4-hydroxy-3-methoxyphenyl)-N-[2-(4-hydroxyphenyl)-2-methoxyethyl]acrylamide (11), perilloside C (12), perilloside A (13), (6S,9R)-9-hydroxy-megastigma-4,7-dien-3-one-9-O-β-D-glucopyranoside (14) and (6S,9R)-roseoside (15) through spectroscopic evidences. The HPLC analysis revealed that hot-water extract of Perillae Folium contained caffeic acid, rosmarinic acid and glycosides of apigenin, luteolin and scutellarein as main constituents.

Five known flavonoid glycosides were isolated from the aerial parts of Lespedeza juncea. Using various spectral data, we found the structures of the isolates to be 6″-O-acetylisovitexin (1), afzelin (2), vitexin (3), isovitexin (4), and, kaempferol- 3-O-robinobioside (5). All compounds were isolated for the first time from this plant.

Mentha arvensis is used traditional medicine to treat various disorders. In the present study, M. arvensis were extracted by the solid-phase microextraction (SPME) method and analyzed by gas chromatograph-mass spectrometry (GCMS). We investigated the protective effects and mechanisms of a M. arvensis extract on scopolamine-induced cognitive and memory impairment. Mice were orally pretreated with a M. arvensis extract or normal saline, and then behavior tests were conducted 30 min after scopolamine injection. The antioxidant capacities were analyzed by free radical scavenging (DPPH and ABTS). Acetylcholinesterase (AChE) activity were also measured using Ellman's method ex vivo test. In behavior tests, percent of spontaneous alteration, escape latency and swimming time in target quadrant were improved by the administration of the M. arvensis extract, which suggests that the M. arvensis extract improves memory function in the scopolamine-treated mice model. In addition, M. arvensis extract showed inhibition of the free radical and AChE activity. The results of the present study suggest that the M. arvensis extract ameliorates scopolamine-induced cognitive and memory deficits through the inhibition of free radicals and AChE activity. Therefore, M. arvensis may be a promising neuroprotective agent for management of learning and memory improvements in human dementia patients.

In this study, we shortly introduced the HPLC method validation guideline for the analysis of functional food which was released from the Ministry of Food and Drug Safety of Korea in Dec 2018. The HPLC method validation was performed through the aforementioned HPLC method validation guideline in order to quantitate scopoletin content from the hot-water extract powder of Artemisia annua Linne. The HPLC method was validated by evaluating specificity, accuracy, precision, limit of quantitation and linearity. All parameters were in the suitable ranges which are designated in the guideline, which indicated the current HPLC method is reliable to quantitate the scopoletin content from the hot-water extract of A. annua.

For toxicological evaluation, water extracts and powder from Polygoni Multiflori Radix were made and the component analysis was followed by the Korean Pharmacopoeia method. To verify the stability of the water extract and powder from Polygoni Multiflori Radix used for toxic testing, the stability test was examined after storage at room temperature and in the cold room for one year. Water extract and powder from Polygoni Multiflori Radix were found to be stable for one year. Therefore, the use of the specimen of Polygoni Multiflori Radix after preparation during the animal test turned out to be stable.
