The stem cell therapy in veterinary medicine continues to grow both experimentally and clinically. Application of adipose-derived mesenchymal stem cell (MSC), umbilical cord blood-derived MSC, bone marrow-derived MSC and skeletal muscle-derived MSC for treatment of various diseases are currently in use for several species. However, the differential efficacies of various approaches are still being investigated. In stem cell therapy, common disease animal models include mouse, rat, rabbit, swine, canine, caprine, equine and dolphin are used. Whereas for clinical models, canine, equine are dolphin are used. We need attention and participation from the researchers of tissue engineering and regenerative medicine, and now it is necessary to develop mutual understanding and cooperation between veterinary and human medicine fields. Considering the steady increase in life expectancy slowing or reversing the deleterious effects of aging has garnered considerable interest. Accordingly, functional cosmetics based on stem cell technology (Stem cell cosmetics), which combine the anti-aging concept with high technology, is an emerging trend in the cosmetics industry. Stem cells possess self-renewal properties and the potency to differentiate. Therefore, stem cells are the most important cells in the skin, as they are the source for continuous regeneration of the epidermis. Stem cell cosmetics are developed based on stem cell technology, which involves using extracts or culture media of stem cells. However, cosmetics containing stem cells or their extracts have not been released into the market due to legal, ethical, and safety concerns. Meanwhile, plant stem cells, which circumvent these problems, are highly regarded in the cosmetics industry for improving culture technology. The European Union prohibits the use of cells, tissues, or products of human origin in cosmetics, whereas the Korea Food and Drug Association has allowed the use of sources originating from stem cell media in cosmetics since 2009. The global cosmetics market is worth more than 242 billion dollars; however, cosmetic companies across the world that are developing and launching stem cell cosmetics based on stem cell activators, culture extracts, and culture media are much more focused on the Korean cosmetic market. In the development of novel tissue engineering and regenerative medicine techniques in humans, preclinical studies using laboratory animal models are required to examine the pathogenic mechanisms and the potential treatment of diseases for the clinics and an indispensable step for determination of their safety and efficacy. Regrettably, the etiological pathology of diseases of spontaneous diseases may not be exactly the same despite the similarities in signs and symptoms observed in artificial laboratory animal models. Therefore, the artificial laboratory animal model is not fully suitable for studies on specific target diseases. However, the animals, especially companion animal, that have spontaneous and frequently occurring diseases due to genetic and environmental factors mostly likely mimic the naive pathology of diseases in humans. Consequently, these species can be better subjects of preclinical studies and veterinary clinical trials in order to possibly gauge the effectiveness of the therapeutic strategies in the clinical stage. Hence, research on the spontaneous and frequently diagnosed diseases of animals predisposed by the type of breeding, age, and gender that are applicable in tissue engineering and regenerative medicine could be an important data for future advancements. In this study, we conducted a survey on the spontaneous and common diseases of canine and feline species to provide a foundation on the effectiveness and practical application of clinical studies. This article overall describes the development of histological method especially, cartilage and bone research in regenerative medicine. Kawamoto's film method, which has been recently introduced into our laboratory, has been applied in various fields that make sectioning of hard tissues easier than the conventional method. Moreover, this method also does not require the time-consuming chemical fixation and/or decalcification process. Kawamoto method involves the use of the adhesive plastic film instead of a cover glass where the thin tissue sections are attached efficiently at low temperatures (-25°C). Furthermore, the histological method preserves the enzymatic activity in the fresh sections in comparison to that of chemically-treated tissue sections. In fact, we used this Kawamoto's film method in one of our researches in which drug repositioning was employed for cartilage regeneration. Some of the Kawamoto-processed tissue sections are featured in this review article. Therefore, the application of this tissue preparation technique allowed effective and histological and histochemical studies within a shorter preparation time with ease and convenience. In the future, this Kawamoto method may offer further applications in the preparation of a more diverse tissues and samples not only in the preclinical but also in the clinical studies. Biomechanical characteristics of articular cartilage in activities of daily life are essential factors to develop tissue-engineered cartilage as a substitution of defect lesion of articular cartilage. However, few studies have explicitly addressed biomechanical characteristics of tissue-engineered cartilage having inferior mechanical material properties. The purpose of this study was to estimate biomechanical characteristics of tissue-engineered cartilage induced in the tibiofemoral joint under compressive load condition using finite element analysis. The two-dimensional axisymmetric finite element model of human tibio-femoral joint was made and five set of partial mechanical properties of tissue-engineered cartilage were applied: Intact (Mo), up 50% (M1.5), down 50% (M0.5), down 90% (M0.1), and down 99.9% (Mx). The results of our study showed that over 50% down in material properties of tissue-engineered cartilage at knee joint were averagely recommended compared to the intact by considering calculated stresses at the contact area between femoral articular cartilage and meniscus. These results could be used as basic data to determine the appropriate mechanical property of the tissue-engineered cartilage for cartilage regeneration. Cornus officinalis has been widely used as herbal medicine exerting various pharmacological actions such as diuretic, anti-allergic, hypotensive and antibiotic activities. Thus we studied the effect of Cornus officinalis on the proliferation, differentiation and matrix mineralization of MC3T3-E1 osteoblast-like cell line. To do the experiments, Cornus officinalis extract (COE) was prepared using ethanol at different concentrations. The cytotoxic effect and the osteogenic differentiation were examined using MTT assay, alkaline phosphatase (ALP) activity, ALP staining, Alizarin red S staining and reverse transcriptase-polymerase chain reaction for osteogenic differentiation markers such as collagen type I (Col-I), ostoecalcin (OCN). The results indicated that there was no effect on cell proliferation on both 1st and 3rd days after treatment in MC3T-E1 cells. The level of ALP, Col-I mRNA, OCN mRNA and mineralized nodule were significantly higher in COE treated group than no treated group. Our results suggest that the COE promotes the osteoblast differentiation and mineralization without observable cytotoxicity. Therefore, it may play an important role in osteoblastic bone formation, and may possibly to be an effective measure for bone regeneration. Tissue Eng Regen Med 2015;12(Suppl 2):113-121

This study was conducted to assess stabilities of the human urine derived stem cells (USCs) during serial subcultures to demonstrate clinical feasibility for future stem cell therapy. For this work, we compared to cell morphology, proliferation rate, immunophenotype, multi- lineage differentiation potentials and immunomodulatory effect of USCs. USCs maintained cell morphology, cell proliferation ability through serial culture (till passage 7). In immunophenotypical analysis, USCs revealed positive for embryonic/mesenchymal stem cell marker (stage specific embryonic antigen, SSEA4) and mesenchymal stem cell markers (CD44, CD73, CD90 and CD105), while negative for hematopoietic stem cell markers (CD34, CD45 and C-kit) and immunogenic marker (HLA-DR). The USC had multi-differentiation potencies into adipocytes and osteocytes, and these abilities maintained into passage 7. The immunomodulatory effect of USCs with phytohemagglutinin stimulated peripheral blood mononuclear cell proliferation was analyzed with two systems, co-culture or separated culture system. The peripheral blood mononuclear cells' proliferation was effectively inhibited on both systems, and the value was more effective with co-culture system than separate one. The present study suggests that sequential sub-passages till passage 7 could maintain stem cell characteristics, multipotent properties, and immune suppressive effect of USCs which should be considered in their therapeutic application in regenerative medicine. Human embryonic stem cells are multipotent cells. In this study, We observed osteogenesis of human embryonic stem cell derived mesenchymal stem cells. mbryonic body formation method was used to derive mesenchymal stem cells from human embryonic stem cells. Embryonic stem cell derived mesenchymal stem cells were immunostained for CD 73 to make characterization of mesencymal stem cells. Osteogenesis of CD 73 positive mesencymal stem cells with media included ascorbic acid, dexamethasone and glycerophosphate was induced. After 10 days culture with osteogenesis media, the cells were immunostained with type I collagen, osteocalcin and Runx-2. After 21 days culture with osteogenesis media, the cells were stained with alizarin red to observe bone nodule formation. Mesenchymal stem cells were derived from human embryonic stem cells by embryonic body method. After these cells were cultured with osteogenesis media, the cells were differentiated into osteoblast and showed bone nodule formation. Tissue Eng Regen Med 2015;12(Suppl 2):132-137

The purpose of this study was to evaluate and compare the potential of using various biomaterials as a carrier of rhBMP-2 in the subcutaneous layer of rats. 5 subcutaneous defects were made in each of the dorsal surfaces of 10 rats. Then, the rats were divided into the following groups: Control group: sham-surgery control, Experimental group 1: rhBMP-2/Bio-Oss®, Experimental group 2: rhBMP-2/Collatape®, Experimental group 3: rhBMP-2/nCS. Histological and histometrical analysis were performed after the rats were sacrificed at 2 and 4 weeks. Regardless of the type of carrier, the groups treated with rhBMP-2 showed significantly greater bone formation when compared to the control group. The amount of new bone formation was greater at 4 week than 2 week. Total bone formation (%) at 4 week was the greatest in rhBMP/Bio-Oss® (26.76±10.13) group, followed by rhBMP/Collatape® (20.38±5.27) and rhBMP/nCS (4.34±2.58). Within the limitation of this study, the results showed that Bio-Oss®, Collatape®, nanocalcium sulfate all showed new bone formation when used as a carrier of rhBMP-2 in the subcutaneous layer of rats, indicating potential use as carriers for rhBMP-2. Fetal cartilage-derived progenitor cells (FCPCs) are a novel cell source for tissue engineering and cell therapy. Previously, cells from various fetal tissues were shown to have immune-modulatory activity just like mesenchymal stem cells. In this study, we investigated the cytotoxicity and therapeutic possibility of FCPCs as a potential cell therapy for arthritis in vivo. To evaluate the acute toxicity and safety, FCPCs were labeled with PKH-26 and subjected to intra-articular injection in rats. When examined at 1 and 2 weeks, the PKH-26 labeled FCPCs were observed only in the synovial membrane but not in other organs. There were no changes either in the white blood cells, red blood cells, and platelets in the hematological analysis and in the weight of the internal organs at 1 week. Therapeutic effect of FCPCs was examined on the complete Freund's adjuvant (CFA)-induced knee arthritis in rats in comparison with triamcinolone, a representative anti-arthritis drug. When inflammation-related edema of the arthritic knee joint was measured by the knee circumference, it increased significantly from the beginning and maintained until the end of measurement in the CFA group, which decreased back rapidly from 1 day in the triamcinolone group and slowly after 7 days in the FCPCs group. In the histological analysis, both triamcinolone and FCPCs decreased the infiltration of neutrophils and lymphocytes into the arthritic synovial membrane gradually at 3 and 7 days. These results suggest that FCPCs shows low toxicity or immune rejection when injected in the synovial cavity and have a therapeutic potential on the synovial arthritis in the rat model.
