1울릉도 수집 균주의 배양조건에 따른 균사 생장 특성 비교

The collection of biological data of indigenous species must comply with the Nagoya Protocol. Fungi contain various bioactive substances making them an attractive source of several products, including food and medicines. In this study, we investigated the growth characteristics of five indigenous fungal strains, Fomitiporia punctata, Polyporus ulleungus, P. brumalis, Gymnopus subnudus, and Tyromyces kmetii, isolated from samples collected in the Ulleungdo Island. The growth rates for each strain were assessed across various temperatures (20 ºC to 35 ºC), culture media (Potato dextrose agar, Malt extract & Yeast extract agar, Malt extract agar, Malt extract & peptone agar, Sabouraud dextrose agar, and Modified Melin-Norkrans agar), and pH conditions (4.0 to 8.0). Additionally, we assessed the mycelial growth characteristics in liquid culture. The mycelial growth in different media varied across species; specifically, F. punctata (in MMNA), G subnudus (in MMNA), and P. brumalis (in MEPA) showed rapid growth. Optimal growth temperatures ranged between 25 ºC and 30 ºC for most species, with the exception of T. kmetii and P. brumalis, which were able to grow across all the temperatures tested. P. brumalis showed the best growth rate, whereas P. ulleungus showed the lowest growth potential. The optimal pH conditions for mycelial growth ranged between 4.0 and 5.0. In experiments using culture flasks, the dry weight of the culture filtrates decreased with the increasing incubation time and showed a significant decrease between 1 and 6 months of incubation, indicating that the five strains take longer than a month to fully use the culture media. Our findings highlight and establish the optimal growth conditions for five different fungal species that can be used in future application studies.

2A New Report of Biscogniauxia petrensis Isolated from Mosquitoes in Korea

A fungal strain designated KNU-WDM2A2 was isolated from mosquitoes in Gimcheon, Korea. The pure culture was transferred to potato dextrose agar (PDA) and synthetic nutrient agar (SNA) media and attained a diameter of 90 mm after 10 days of incubation at 25℃. The colonies were whitish to light pink and cottony to wooly, with an abundant production of aerial mycelia. The strain produced hyaline to slightly yellowish conidiophores that were rough-walled and branched, with conidiogenous cells arising terminally or laterally. Conidia were unicellular, hyaline to light brown, smooth, and oval or ovoid to clavate, with a size of 4.1-6.9×2.5-3.3 μm (n=65). A phylogenetic analysis was conducted using the internal transcribed spacer (ITS) regions and 28S rDNA of large subunit (LSU) sequences, to support the cultural and morphological characteristics. The KNU-WDM2A2 strain was identified here as Biscogniauxia petrensis, new to Korea.

This study was carried out to develop a new method of cultivation of 'Hwadam' a new variety of Lentinula edodes, to improve fruiting body productivity and reduce labor. To determine the optimum filter number for box cultivation, L. edodes were assessed for their cultural characteristics and mushroom yield. During spawn running of L. edodes in the box, the temperature gradually increased to the highest value of 25.8℃, 23.7℃, and 23.0℃ on day 18 for the four filter, two filter, and no treatment groups, respectively. Oxygen concentration displayed an opposite trend to carbon dioxide concentration, reaching the lowest value on day 21. The oxygen concentration progressively increased as more filters were used (5.4%, 8.2%, and 8.9% treatment with no, two, and four filters, respectively. Growth of L, edodes in the box resulted in the highest yield (2,228 g/7kg substrate) and biological efficiency (70.7%) using four filters. The biological efficiency of the four filter treatment was 29.4% higher compared to bag cultivation (41.3%). Further studies are necessary to verify stable productivity of fruiting bodies through repeated cultivation.

4First report of six Sordariomycetes fungi isolated from plant litter in freshwater ecosystems of Korea

Freshwater ecosystems provide a complex environment for microorganisms. In this study, we isolated diverse fungal strains from plant litter in freshwaters. These strains were identified using molecular phylogenetic analyses of rDNA and/or other gene sequences (TUB, GAPDH, and EF1 ). In addition, we examined their morphological characteristics by microscopy and cultural characteristics on several media. We identified six previously unrecorded Sordariomycetes species in Korea, i.e., Colletotrichum godetiae, Discosia rubi, Robillarda sessilis , Monochaetia dimorphospora , Idriella lunata , and Phialemoniopsis endophytica . Of these, D. rubi and M. dimorphospora exhibited high extracellular amylase, lipase, and protease activities, suggesting that these fungal isolates might play an important role as decomposers in freshwater ecosystems. Plant litter could thus be a good source for isolating and investigating previously undocumented fungal species in freshwater environments.

In this study, we isolated endophytic fungi from the conifer leaves of Pinus densiflora inhabiting Mt. Gyehwasan in Korea. We identified the isolated fungal strains based on their morphological characteristics and phylogenetic analysis using DNA sequences of the internal transcribed spacer, small subunit rDNA, large subunit rDNA, and β-tubulin region. In the process of this study, we confirmed two endophytic fungi that have not been recorded previously in Korea, Lophiostoma macrostomum and Pestalotiopsis lespedezae. In this report, we described the morphological characteristics of these fungal strains and the results of their molecular analysis.

A new oak mushroom cultivar 'Hwadam' was bred from monokaryotic strains of 'GMLE36062' and 'LE15401'. The optimum temperature for mycelial growth of 'Hwadam' on potato dextrose agar was 25℃. The cultivation period of 'Hwadam' lasted for 129 days until the first harvest of the fruiting bodies, including 35 days for spawn run, 84 days for browning, and 10 days for development of fruiting bodies. The fruiting bodies of 'Hwadam' had a hemispherical, brighter pileus and thicker stipe than the control cultivar (Sanjo 701ho). The three-cycle yield of 'Hwadam' was 554 g, which was higher than that of control cultivar (405 g). In addition, 'Hwadam' contained 79.4 mg/g of beta-glucan and 0.93 mg GAEs/g of phenolic compounds, which was similar to that of the control cultivar but higher than that of 'L808' and 'Chujae 2ho'.

7A Survey of Macrofungal Diversity in Da-Lat, Southern Vietnam

Da-Lat is a hilly area located in southern Vietnam. Macrofungal diversity of Da-Lat was investigated from 2018 to 2019. A total of 468 macrofungal specimens was collected and identified using the modern species concept and taxonomic and phylogenetic analyses. Among them, internal transcribed spacer(ITS) region of 401 specimens were successfully sequenced and compared with those of related species retrieved from GenBank. In total, 180 specimens were identified at the species level. The sequenced specimens were classified into 2 phyla, 13 orders, 38 families, 93 genera, and 124 species. The remaining 221 specimens (175 species) did not match the species level. This study is the first well-documented taxonomic list of macrofungi collected from southern Vietnam.

The goal of this study was to investigate the diversity present among wild yeasts obtained from soils of spice fields and from mountain soils, and to further, characterize previously unrecorded novel wild yeast strains. In total, 36 strains from 17 different species of wild yeasts were isolated from 35 soil samples obtained from garlic fields of Geumsan, Chungcheongnam-do, Korea. Among these, six yeast strains of Trichosporon moniliiforme, and four strains each of Papiliotrema flavescens and Candida melibiosica species were isolated. Additionally, 22 strains of 18 different species of wild yeasts were isolated from 32 soil samples collected from the ballonflower and ginger fields of Geumsan, Korea. Finally, 46 strains of wild yeasts were isolated from 35 soil samples obtained from Mt. Daedun in Geumsan, Korea. Among the total of 106 isolated wild yeast strains, 10 strains, including Debaryomyces vindobonensis GHY31-3 represented novel yeast strains which were previously unrecorded. All the 10 previously unrecorded yeasts were oval or global in shape, and five strains, including Filobasidium stepposum SFG1-4 formed ascospores. Three strains, including Pseudozyma alboarmeniaca CD 23-5 grew well in vitamin-free medium. Cell-free extract obtained from Filobasidium magnum SFG1-3 indicated 28.6% of xanthine oxidase inhibitory activity.

국내에 자생하는 지렁이로(붉은줄지렁이)의 장에서 야생효모를 분리, 동정하고자 하였다. 지렁이는 고양시 난지 물 재생 센터에서 분양 받았으며, 지렁이 5마리에서 19개의 야생효모를 분리하였다. 효모의 D1/D2 영역으로 동정하여 국내에 보고되지 않은 효모 3균주를 선별하였다. 국내 미기록종 효모인Yarrowia deformans YP242 (=KACC48778), Sporidiobolus pararoseus YP66 (=KCTC27963), Naganishia liquefaciens YI9 (=KACC48948) 균주는 위상차현미경, API 20C AUX kit를 통해 균학적 특성을 조사하였다. YP242, YI9, YP66 균주의 세포 모양은 타원형이며, 집락은 볼록하고, 부드러운 재질을 가지는 것으로 관찰되었다. 탄소원 활성 측정 결과, YP242는 glycerol, L-arabinose, N-acetyl-D-glucosamine을, YP66는N-acetyl-D-glucosamine을, liquefaciens YI9는 2-keto-D-gluconate을 탄소원으로 사용할 수 있음을 확인하였다.


The goal of this study was to isolate and characterize the wild yeast strains from the gut of earthworm(Eisenia andrei). The 19 yeast strains isolated from 5 gut of earthworm samples from Nanji water regeneration center in Goyang-si, Gyeonggi-do, Korea. Among them, 16 strains were recorded and 3 strains, Yarrowia deformans YP242 (=KACC48778), Sporidiobolus pararoseus YP66 (=KCTC27963) and Naganishia liquefaciens YI9 (=KACC48948) were recorded for the first time in Korea. The microbiological characteristics of these previously unrecorded yeasts were investigated. All three strains were oval-shaped, convex and smooth. However, they showed some differences in colony color and result of carbon assimilation assays. YP242 was white-colored and assimilated glycerol, L-arabinose and N-acetyl-D-glucosamine as carbon sources. YP66 was red-colored and assimilated D-Saccharose. YI9 was white-colored and positive for 2-keto-D-gluconate assimilation.

We report on an unrecorded endophytic fungus, Grovesiella abieticola (Zeller & Goodd.) M. Morelet & Gremmen isolated from Abies koreana of Mt. Halla in Jeju. In this study, we used a culture method to determine its conidia and compared relative species using an internal transcribed spacer (ITS) 1, 2, and 5.8S ribosomal DNA sequence. Thus, we present the cultural characteristics and morphology of G. abieticola in this paper.
