1흰목이 균사체 배양 적합 조건 설정

This study was conducted to investigate the optimum culture conditions, for mass production of Tremella fuciformis in M9 basic medium. The strain KMCC04674, used in this study was identified T. fuciformis by internal transcribed spacer (ITS). To define the optimum conditions for the mass production of T. fuciformis, we investigated the effects of different culture conditions and various nutrient sources on the fungal growth. The optimum initial pH and temperature for the fungal growth were 5.0 and 25℃, respectively. The optimal composition of the growth medium was 4.0% mannitol, 3.0% NH4H2PO4, 1.0% malt extract, 1.0% glutamic acid, 5mM CaCl2, and 0.5% glucanic acid.

2First Report of Leptosphaerulina saccharicola Isolated from Persimmon (Diospyros kaki ) Tree Bark in Korea

A fungal strain, designated PTT-2, was isolated from the bark of the trunk of a persimmon (Diospyros kaki) tree in Cheongdo, Korea. The isolate showed morphological similarities with Leptosphaerulina saccharicola. Strain PTT-2 had more rapid growth on potato dextrose agar medium than on oatmeal agar, malt extract agar, and synthetic nutrient poor agar media, with colony sizes of 53.8 mm, 49.8 mm, 48.4 mm, and 28.1 mm after 7 days at 25℃ temperature, respectively. Strain PTT-2 produced ascospores, which had irregular wavy edges, oblong to ellipsoidal shape, hyaline appearance and 23.6 × 10 μm size. The black ascomata were developed on PDA medium, and asci were recorded. A BLAST search of the internal transcribed spacer (ITS) region, TEF1-α and RPB2 gene sequences revealed that strain PTT-2 showed more than 99% nucleotide similarity with a strain of Leptosphaerulina saccharicola previously reported from Thailand. A neighbor-joining phylogenetic tree was constructed by concatenating the above-mentioned sequences, and showed that strain PTT-2 clustered in the same clade with L. saccharicola. Based on these findings, this is the first record of Leptosphaerulina saccharicola occurring in Korea.

We extracted arbuscular mycorrhizal fungal (AMF) spores from rhizospheres of three plants from Upo Wetland, Korea. We identified the isolated AMF spores based on morphological characteristics and phylogenetic analysis of partial 18S rDNA nucleotide sequences. The species diversity of AMF spores was calculated among the study sites and host plants. Consequently, nine species from six genera of AMF spores were identified. We confirmed the species diversity of the AMF spores in rhizospheres affected by host plants in the wetland. In the course of this study, we confirmed a previously unreported AMF species in Korea: Diversispora epigaea. We described the morphological features and molecular characteristics of this previously unreported AMF species.

4First Report of Allantophomopsiella pseudotsugae Isolated from Soil in Korea

A fungal isolate designated 17E029 was isolated from a soil sample in Jeju, Korea. The strain was similar to other Allantophomopsiella species in its morphological characteristics such as grey mycelia, conidiophore, and conidia sizes. The isolate produced aerial mycelia, which appeared grey on the reverse side of the media surfaces and turned black on the front side of the colonies. The conidiophores emanating from the hyphae were hyaline, grey, aseptate, branched, and 6.7~9.2 × 1.8~2.5 μm. Conidiogenous cells were ovoid to subcylindrical, discrete, guttulate, and hyaline. Conidia were hyaline, aseptate, smooth, guttulate, oval to subcylindrical, irregular in shape, and 6.0~7.8 × 3.0~3.4 μm. The strain was confirmed based on phylogenetic analysis of the closest related organism, A. pseudotsugae CBS 288.37, using the partial 28S, internal transcribed spacer rDNA regions, and partial RNA polymerase II second largest subunit locus (RPB 2) gene sequences along with its culture characteristics. Therefore, morphological observations and phylogenetic analysis revealed that strain 17E029 is similar to the previously identified A. pseudotsugae. Hence, this species was described as A. pseudotsugae strain 17E029, which is a new record in Korea.

We isolated endophytic fungi from the conifer leaves of Pinus densiflora inhabiting Buan-gun, Jeollabuk-do, Korea. We identified the isolated fungal strains based on phylogenetic analysis performed using the nucleotide sequences of the internal transcribed spacer, large subunit, and beta-tubulin. We confirmed the presence of three novel endophytic fungi in Korea, namely Paracamarosporium hawaiiense, Tubakia dryina, and Zasmidium fructigenum. In this report, we described the morphological characteristics of these fungal strains and the results of their phylogenetic analysis.

We investigated the biodiversity of endophytic fungi of Bolboschoenus planiculmis and Schoenoplectus triqueter in Korea. Twenty five of these host plants were collected from six sampling sites. Internal transcribed spacer analysis identified a total of eighty five isolates from the collected plants. The isolates comprised nineteen taxa; most of the isolates (92%) belonged to class Dothideomycetes, followed by Sordariomyetes (4.8%), Agaricomycetes (1.6%), and Eurotiomycetes (0.8%). The most prevalent fungus was Cladosporium perangustum in B. planiculmis and Macrospora scirpicola in S. triqueter. There are very few studies of endophytic fungi in halophytes and these have reported biodiversity patterns different from those of previous studies in Korea. Our results should provide useful information for future research in Korea.

The goal of this study was to elucidate wild yeast diversity of Geumgang midstream near Sejong metropolitan autonomous city, Korea. Thirty-seven strains of 32 species of wild yeasts were isolated from 43 water and soil samples under the Bulti bridge of Sejong city, Korea. Seven yeasts of each Candida spp. and Cryptococcus spp. were the predominant species isolated from samples near the Bulti bridge. Holtermanniella takashimae SW048 (NNIBRFG9314), Cystofilobasidium infirmominiatum SW013 (NNIBRFG9310), Mrakia cryoconite SW015 (NNIBRFG9316), Pichia sporocuriosa SW085 (NNIBRFG9326) and Cryptococcus aspenensis SW008 (NNIBRFG9309) represented novel yeast strains found in Korea for the first time. All of these previously unrecorded yeasts, except for Mrakia cryoconite SW015 had ascospores and grew well in yeast extract-peptone-dextrose (YPD), yeast extract-malt extract (YM) and potato-extrose (PD) media. Pichia sporocuriosa SW085 grew well in vitamin-free medium and Holtermanniella takashimae SW048, which was a halotolerant wild yeast, grew well YPD medium containing 5 % NaCl. Twenty-six strains representing eight species of wild yeast were isolated from 22 water and soil samples under the Haetmuri bridge of Sejong city, Korea. Candida pseudolambica (12 strains) and Aureobasidium pullulans (11 strains) were the predominant isolates from samples near the Haetmuri bridge. Occultifur kilbournensis HB060 (NNIBRFG9317), Sampaiozyma vanillica HB014 (NNIBRFG9332), Xenoramularia neerlandica HB039 (NNIBRFG9335), Candida norvegica HB315 (NNIBRFG9306), C. melibiosica HB316 (NNIBRFG9305), C. quercuum GB014 (NNIBRFG9307), and C. succiphila GB015 (NNIBRFG9308) represented novel yeast strains recorded in Korea for the first time. O. kilbournensis HB060 and X. neerlandica HB039 did not form ascospores or pseudo-mycelia. All of these previously unrecorded yeasts, except S. vanillica HB014 and X. neerlandica HB039, grew well in vitamin-free medium, and C. norvegica HB315 and C. succiphila GB015, which were halotolerant wild yeasts, which grew well in YPD medium containing 5 % NaCl.

Mycelial browning, which protects the organism from contamination and moisture loss, is essential for sawdust cultivation of Lentinula edodes. The effects of light and light wavelengths on the mycelial browning of the L. edodes Sanjo 701ho strain, and the characteristics of its brown hyphae, were investigated. After the mycelia were cultured on potato dextrose agar medium under fluorescent lamps covered with colored cellophane filters (red, green, and blue) or under light emitted diodes (LED), with wavelengths ranging from 400 to 700 nm (far-red, red, green, and blue), for 14 h per day for 40 days, the mycelial browning rate was measured. The wavelength of fluorescent lamps, which range from 300 to 1,100 nm, was reduced to 360 to 1,022 nm with the use of three colored cellophane filters and the photosynthetic photon flux density was reduced by 42 to 71 % depending on the light wavelength. The browning rate by colony area of mycelia exposed to light was at an average of 64 %, whereas, that of unexposed mycelia was only 5 %. The browning rate was 0.02 % in far-red, 1.5 % in red, 53.8 % in green, 57.3 % in blue, and 64.0 % in fluorescent light. The white mycelia were resilient with actively growing hyphae, filled with cytoplasm, and thin cell walls less than 1 μm thick. Conversely, the brown mycelia possessed dead, hard hyphal structures without cytoplasm, but with approximately 2-4 μm-thick cell walls. In conclusion, lights of varying wavelengths, especially short-wavelength LEDs, are effective for forming dead, brown mycelia of L. edodes, thus, forming a protective functional layer for its living white mycelia.

The goal of this study was to characterize unrecorded wild yeasts from soils of spices plants fields and further, to elucidate its anti-demential activities and tyrosinase inhibitory activity. Piskurozyma taiwanensis R4-1 (NIBRFGC000502619), Nadsonia starkeyi-henricii R6-2 (NIBRFGC000502618), and Canadida friedrichii M12-6 (NIBRFGC000502615) isolated from soil of garlic field represented newly recorded yeast strains in Korea. Vishniacozyma peneaus I2-9 (NIBRFGC000502617) and Cryptococcus aspenensis I21-1 (NIBRFGC000502616) from soil of ginger field represented also newly recorded yeast strains, and microbiological characteristics of its fifteen yeast strains were investigated. All of these unrecorded yeasts exhibited oval-global shape and have ascospores except Canadida friedrichii M12-6. Piskurozyma taiwanensis R4-1 and Canadida friedrichii M12-6 grew well in vitamin-free medium, and Piskurozyma taiwanensis R4-1 was halotolent growing in 10% NaCl-containing yeast extract peptone dextrose (YPD) broth. After prepared cell-free of the unrecorded wild yeasts, acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) inhibitory activities as anti-dementia activity and tyrosinase inhibitory activity as whitening activity were determined. Cell-free extract from Canadida friedrichii M12-6 had the highest tyrosinase inhibitory activity of 14.4%.

A previous record of Coleosporium phellodendri on Zanthoxylum ailanthoides in Korea by Korean researchers was regarded as C. zanthoxyli by foreign scientists without further studies. To clarify the identity of a Coleosporium species occurring on Z. ailanthoides in Korea, we examined the morphological characteristics of eight materials and analyzed the internal transcribed spacer (ITS) and 28S large subunit (LSU) rDNA regions of three representative specimens which were collected in Korea and deposited in the Mycological Herbarium at Korea University (Seoul, Korea). All specimens collected in Korea were morphologically and molecularly determined to be C. zanthoxyli. Therefore, this is the first confirmed report on the presence of C. zanthoxyli on Z. ailanthoides in Korea.
