1Effects of enzymatically hydrolyzed fish byproducts in diet of juvenile rainbow trout (Oncorhynchus mykiss)

Five experimental diets were formulated to evaluate the effects of dietary enzymatically hydrolyzed tuna by-product on growth, non-specific immune responses, and hematology of juvenile rainbow trout (Oncorhynchus mykiss). A basal diet with 50% of fishmeal was used as control (CON) and four other diets replaced 12.5% (TBB12.5), 25% (TBB25), 37.5% (TBB37.5), and 50% (TBB50) of fish meal in the CON diet. Juvenile rainbow trout (4.87 ± 0.05 g) were randomly distributed into 15 tanks (50 L) and fed 3-4% of wet body weight two times a day. At the end of 7 weeks of feeding trial, weight gain, specific growth rate, feed efficiency, and protein efficiency ratio of fish fed CON diet were significantly higher than those of fish fed TB50 diet (P < 0.05). But there were no significant differences among fish fed CON, TBB12.5, TBB25, and TBB37.5 diets (P > 0.05). There were no significant differences in GPT levels among fish fed CON, TBB12.5, TBB25, and TBB37.5 diets. Also, there were no significant differences in lysozyme, superoxide dismutase, glucose, and total protein levels in all experimental diet (P > 0.05). The broken-line analysis indicated that the minimum dietary level of enzymatically hydrolyzed tuna by-product to replace fishmeal could be 29.7% in rainbow trout. These results indicated that the optimum level of dietary enzymatically hydrolyzed tuna by-product could replace greater than 29.7% but less than 37.5% of fishmeal in juvenile rainbow trout diet.

In the present study, the protective effects of Hizikia fusiforme and Chlorella sp. extracts on lead acetate-induced hepatotoxicity were investigated. Hepatic damage was induced in rats by intraperitoneal (i.p.) injection of lead acetate and the protective effects of H. fusiforme (HZK) and Chlorella sp. (CHL) extracts on lead acetate-induced hepatic damage in rat liver were examined. The results revealed significantly increased glutamic oxaloacetate and glutamic pyruvic transaminase levels in the group treated with lead acetate only (Pb group); oral administration of HZK and CHL extracts tended to decrease the enzyme levels similar to those observed in the control group. Regarding antioxidant enzymes, superoxide dismutase activity was increased in the Pb group and decreased in a concentration-dependent manner in the HZK- and CHL-treated groups. Glutathione levels were increased in a concentration-dependent manner in the HZK- and CHL-treated groups. There was no significant difference in catalase activity. Western blot analysis showed inflammation-related protein expression in mitogen-activated protein kinase and Nrf2 pathways was affected in the HZK- and CHL-treated groups. Therefore, HZK and CHL extracts exerted antioxidant and anti-inflammatory effects against lead acetate-induced hepatotoxicity. Development of functional health foods containing HZK and CHL extracts, which have hepatoprotective effects against inhaled lead acetate, should be considered.

3Toxic effects of arsenic on growth, hematological parameters, and plasma components of starry flounder, Platichthys stellatus, at two water temperature conditions

The purpose of this study is to investigate the changes in growth, hematological parameters, and plasma components of juvenile starry flounder, Platichthys stellatus, following exposure to varying arsenic concentrations present at different water temperatures. P. stellatus (total length, 15.9 ± 0.4 cm; body weight, 62.2 ± 4.2 g) were exposed for 4 weeks to waterborne arsenic (sodium arsenite, As) at 0, 150, 300, and 600 μg/L at temperatures of 12 °C and 18 °C. Toxic effects of As exposure on P. stellatus were higher at the higher temperature and the growth and hematological parameters measured decreased with increasing arsenic concentration, while the concentration of plasma components measured increased. This indicates that waterborne As exposure and water temperature can cause toxic effects on growth, hematological parameters, and plasma components in Platichthys stellatus.

4Ethanol extract separated from Sargassum horneri (Turner) abate LPS-induced inflammation in RAW 264.7 macrophages

Background: This study is aimed at identifying the anti-inflammatory properties of 70% ethanol extract produced from an edible brown seaweed Sargassum horneri (SJB-SHE) with industrial-scale production by Seojin Biotech Co. Ltd. S. horneri is a rich source of nutrient and abundantly growing along the shores of Jeju, South Korea.
Methods: Here, we investigated the effect of SJB-SHE on LPS-activated RAW 264.7 macrophages. The cytotoxicity and NO production of SJB-SHE were evaluated using MTT and Griess assays, respectively. Additionally, protein expression and gene expression levels were quantified using ELISA, Western blots, and RT-qPCR.
Results: Our results indicated that pre-treatment of RAW 264.7 macrophages with SJB-SHE significantly inhibited LPS-induced NO and PGE2 production. SJB-SHE downregulated the proteins and genes expression of LPS-induced iNOS and COX2. Additionally, SJB-SHE downregulated LPS-induced production of pro-inflammatory cytokines (tumor necrosis factor-α, interleukin (IL)-6, and IL-1β). Furthermore, SJB-SHE inhibited nuclear factor kappa-B (NF-κB) activation and translocation to the nucleus. SJB-SHE also suppressed the phosphorylation of mitogen-activated protein kinases (ERK1/2 and JNK).
Conclusions: Collectively, our results demonstrated that SJB-SHE has a potential anti-inflammatory property to use as a functional food ingredient in the future.

5Hexane fraction from the ethanolic extract of Sargassum serratifolium suppresses cell adhesion molecules via regulation of NF-κB and Nrf2 pathway in human umbilical vein endothelial cells

Sargassum serratifolium ethanolic extract has been known for strong antioxidant and anti-inflammatory properties. We prepared hexane fraction from the ethanolic extract of S. serratifolium (HSS) to improve biological activities. In this study, we investigated the effects of HSS on the inhibition of tumor necrosis factor (TNF)-α-induced monocyte adhesion to human umbilical vein endothelial cells (HUVECs). We found that HSS suppressed the production of cell adhesion molecules such as intracellular adhesion molecule-1 and vascular cell adhesion molecule-1 in TNF-α-induced HUVECs. Moreover, TNF-α-induced production of monocyte chemoattractant protein 1 and keratinocyte chemoattractant was inhibited by HSS treatment. HSS suppressed TNF-α-induced nuclear factor kappa B (NF-κB) activation via preventing proteolytic degradation of inhibitor κB-α. HSS induced the production of heme oxygenase 1 via translocation of Nrf2 into the nucleus in TNF-α-treated HUVECs. Overall, HSS alleviated vascular inflammation through the downregulation of NF-κB activation and the upregulation of Nrf2 activation in TNF-α-induced HUVECs. These results indicate that HSS may be used as therapeutic agents for vascular inflammatory disorders.

Russian sturgeon (Acipenser gueldenstaedtii) is commercially important in Korea because its caviar is highly prized. Although the early ontogeny of the species has been described, behavioral modifications in response to various light intensities or diel photoperiodicity patterns have not been studied extensively. The objective of the present study was to examine the behavioral characteristics of hatchery-produced A. gueldenstaedtii prelarvae over a diel photoperiodic cycle. During a diel light cycle comprising 16 h of daylight (450 lx), 4 h of dim light (10 lx), and 4 h of darkness (< 1 lx), newly hatched A. gueldenstaedtii prelarvae exhibited negative phototaxis in daylight (day 0-day 2), and this early behavior was not significantly affected by changes of light intensities. Rheotactic and non-rheotactic aggregation into schools was typical between days 3 and 6. Under the diel light cycle conditions described, rheotaxis was not influenced by environmental light intensity as much as non-rheotactic schooling behavior. Post-schooling behavior, which progressed from day 7 to day 8, did not change significantly during the diel light cycle. The data from the present study could be of practical value in developing a visual guide for assessing the fitness and quality of Russian sturgeon prelarvae under diel light cycle conditions in hatcheries.

7Introduction of bacterial and viral pathogens from imported ornamental finfish in South Korea

Background: Live fish import may lead to the unintended introduction of pathogens. We examined the monthly distribution of microbial pathogens in ornamental finfish imported into South Korea over a 6-month period.
Results: Vibrio alginolyticus was detected in one lemon damsel in June and July; V. vulnificus was detected in one lemon damsel, one caerulean damsel, and one pearl-spot chromis and one ocellaris clownfish in July, April, and May, respectively; Photobacterium damselae was detected in one ocellaris clownfish and one caerulean damsel in June and July, respectively; V. anguillarum was detected in one pearl-spot chromis in February; V. harveyi was detected in one ocellaris clownfish and two mandarin fish in February and April, respectively; Yersinia ruckeri was detected in a pearlscale goldfish group in June and July and in two colored carp groups in July; and Lactococcus garvieae was detected in a lemon damsel group and a sutchi catfish group in July and May, respectively. European catfish virus, the only viral pathogen detected, was found in two sutchi catfish groups in May.
Conclusion: This study is the first to identify pathogenic species and the presence or absence of pathogens (nonquarantine diseases) in imported ornamental finfish. These results demonstrate that various pathogens with the potential to harm indigenous fish populations can accompany ornamental finfish imported into South Korea.
